The existance of blood-epididymis barrier has been confirmed. The Ig classification of some monoclonal human sperm and mouse sperm antibodies (Mabs) were determined by immunodiffusion. The mature male mice were passively immunized with these Mabs, HS11, HS63, MS204, MS207 (IgG_1) and MS79, MS212 (IgM) by intraperitoneal injections. Sera and sperm were recovered from the immunized mice and assayed by immunofluorescenceo It was found that all Mabs could enter the blood circulation but were...

Sera and sperm were recovered from the immunized mice and assayed by immunofluorescenceo It was found that all Mabs could enter the blood circulation but were blocked by blood-testis barrier. IgG Mabs could combine with some relative sperm antigens within the different segments of epididymis and IgM could not. It was suggested that there was a barrier to the passage of IgM from serum to the lumen of the mouse epididymis but the barrier did not exclude IgG_1.

用琼脂糖双扩散法测得抗人精子单克隆抗体HS11、HS63和抗小鼠精子单抗MS204,MS207属IgG_1,抗小鼠精子单抗MS79、MS212属IgM。将上述单抗通过腹腔注射被动免疫正常CD-1雄性小鼠,然后收集受鼠血清及其睾丸、附睾各段的精子作免疫荧光染色。发现所有抗精子单抗均能进入血循环,但被血-睾屏障所阻挡。HS11、HS63、MS204和MS207能进入附睾管腔,与相应的精子抗原发生免疫反应,但染色百分率较低。MS79、MS212与附睾各段精子均无染色反应。提示血-附睾屏障能阻止IgM,但不排斥IgG_1。

Results: Strong positive reactions were observed in the free margins of the epithelial cells on the epididymal duct of the head and positive in the basement membrane of the tail, but negative reactions were observed in the body. The results showed that the distribution of ALP in the above three sites of epididymis was distinctly different. Conclusion: The location of ALP in the head, body and tail of the epididymis is distinctly different, suggesting that ALP has different metabolic functions in each site of epididymis during the sperm development and differentiation. We may also assume that ALP involves in the formation of the blood-epididymis barrier.

Lipocalin type prostaglandin D synthase (L PGDS) is widely expressed in male reproductive system, and the extent of its expression is related to the development and maturation of male reproductive organs. The synthesis of L PGDS in epididymis is possibly regulated by testosterone and some factors secreted by testis, while the expression of L PGDS in testis is affected by local spermatogenic cells. The L PGDS which catalyzes prostaglandin D 2 (PGD 2) production in seminal plasma is a kind of glycoprotein with multiple patterns in male reproductive system. L PGDS is a fertility associated protein and related to the transportation of retinoid and substances which transfer through blood testis or blood epididymis barrier. Furthermore, PGD 2 is related to not only the transfer of sperm through female reproductive ducts but also immunity associated infertility. Cloning and expression of human testis L PGDS in vitro are very important to further exploration of L PGDS function in male reproductive system.
