Objective:The aim of this experiment was to observe the consecutive alterations of serum lipid peroxides and cellularities of BALF,so as to study the pathogenesis of pulmonary fibrosis.Methods:Bleomycin was given to mice via a single intratracheal injection to establish animal models of lung fibrosis.Sterile saline was injected intratracheally as control.At days 3,7,14 and 28 after injection,animals were killed,sLPO determination,cellular analysis of BALF and histologic studies were carried out.Results:The contents of sLPO were increased significantly on day 3,peaked on day 7 after bleomycin injection and droped sequentially after that.Simultaneously,total of cell,percentages of neutrophil,lymphocyte and eosinophil were also increased significantly,and their alterations correlated well with that of sLPO.Pathologic histology indicated acute alveolitis between 3 and 14 days after bleomycin injection and diffuse pulmonary fibrosis on day 28. Conclusion:Peroxidation damage and inflammatory cells might play an important role in the development of pulmonary fibrosis.

目的:观察实验性肺纤维化过程中血清脂质过氧化物(sLPO)和支气管肺泡灌洗液(BALF)细胞学的动态变化,以探讨肺纤维化的发病机理。方法:以博莱霉素A5气管内一次注入法复制小白鼠肺纤维化模型,以生理盐水气管内注入作为对照,于注药后第3、7、14、28天分四批处死动物,进行sLPO测定(TBA比色法)、BALF细胞计数、分类及肺组织病理学检查。结果:气管内注入博莱霉素A5后第3天sLPO即显著升高,于第7天达高峰,而后渐降至正常;BALF中白细胞总数、中性粒细胞比例、淋巴细胞比例及嗜酸性粒细胞比例亦显著增高,其变化与sLPO变化相一致;肺组织病理学第3~7天表现为急性肺泡炎,第28天肺纤维化形成。结论:脂质过氧化及炎症细胞在肺纤维化的发生中起重要作用。

Methods Bleomycin A5 was given to mice via a single intratracheal injection to establish the animal model of alveolitis. Sterile saline was injected intratracheally as control. In treatment group, the mice received about 2mg Captopril by lavage everyday after intratracheal injection. At the 7th day after injection, the animals were killed and lung coefficients were calculated, the contents of serum lipid peroxide (sLPO) and the protein contents in BALF were determined, inflammatory cells in BALF were counted, and histologic studies were carried out. Results In the model group, the lung coefficients, the sLPO and protein contents in BALF, the total number of cells, percentages of neutrophils, lymphocytes and eosinophils in the BALF were all found increased significantly. Histopathology revealed acute alveolitis. All these parameters declined significantly after Captopril treatment except the proportion of lymphocytes. Pathologic studies showed alleviation of alveolas inflammation. Conclusion Captopril may have some protective effect on Bleomycin A5 induced alveolitis. (Shanghai Med J, 1999,22∶684 686)

目的 观察卡托普利对平阳霉素致小鼠肺泡炎的影响。方法 以平阳霉素气管内一次注入法造成小鼠的急性肺泡炎模型,治疗组气管内注入平阳霉素后每日灌入卡托普利,进行肺系数、血清脂质过氧化物(sLPO) 和支气管肺泡灌洗液(BALF) 中蛋白含量测定、BALF细胞计数和分类,以及肺组织病理学检查。结果 未治疗组肺系数、sLPO、BALF 中蛋白含量、BALF 细胞总数、中性粒细胞、淋巴细胞和嗜酸性粒细胞比例均较对照组显著升高,肺组织学表现为急性肺泡炎;卡托普利治疗组上述各指标( 除淋巴细胞比例外) 均显著下降,肺组织学显示炎症明显减轻。结论 卡托普利对平阳霉素所致的肺泡炎具有一定的保护作用。
