AIM To study the pharmacokinetics of the new tumor radiosensitizer glycodidazolum natrium(CMNa) in tumor patients after single and multiple dose, in order to evaluate the accumulation of CMNa in patients. METHODS 5 tumor patients were given an intravenous dose of CMNa 700 mg/m 2 every other day for 9 times, and blood samples were collected at designed time after the first and the ninth CMNa administration. The blood concentrations of CMNa were determined by HPLC UV. The blood concentration time curves of CMNa after single dose and multiple dose were fitted to a two compartment open model. RESULTS CMNa was found to be distributed and eliminated rapidly in patients whether after single or after multiple dose. The blood concentration time curves and pharmacokinetics parameters between single dose and multiple dose were similar, and they showed no significant difference. CONCLUSION CMNa will not accumulate in patients′ bodies, and it is a safe drug.

AIM: To evaluate safety of glycodidazolum natrium (GN) by determining GN and metronidazole (Met) elimination rate of tumor radiosensitizer GN after different doses. METHODS: Twentyone tumor patients, separated into 5 dosegroups randomly, were respectively administered 500, 600, 700, 800, 900 mg/m2 GN by iv, and urine samples were collected after administration between designed time interval, and concentration of GN and its metabolite Met in urine was determined by HPLCUV, and elimination rates of 5 dosegroups were calculated. RESULTS: GN mainly eliminated from kidney in patients' bodies, and it mainly eliminated in the form of GN itself. In 5 dosegroups, GN elimination rates in urine were 7433%, 6547%, 5376%, 5804% and 5479%, respectively; and eliminiation rates of metabolite Met were respectively 457%, 390%, 511%, 516% and 374%. GN eliminated rapidly in patients' bodies, in 4 h after administration with more than 95% GN and Met of total elimination quantity. CONCLUSION: It will not store up in patients' bodies, and it is a safe drug.

目的 :测定了放疗增敏药甘氨双唑钠 ( GN)及代谢产物甲硝唑 ( Met)在尿中的排泄 ,评价 GN的安全性。方法 :2 1名肿瘤病人 ,随机分成 5组 ,分别 iv gtt5 0 0、6 0 0、70 0、80 0、90 0 m g/m2的 GN,于滴后设定时间段收集尿样 ,用高效液相色谱 -二极管阵列紫外法测定受试病人尿样中的 GN及代谢产物 Met的浓度 ,并计算出 5个剂量组的平均排泄率。结果 :GN在病人体内以原药肾排泄为主。各剂量组尿中 GN的排泄率分别为 74.33%、6 5 .47%、5 3.76 %、5 8.0 4%、5 4.79% ,而代谢产物 Met的排泄率分别为 4.5 7%、3.90 %、5 .11%、5 .16 %、3.74%。 GN在病人体内消除很快 ,5个剂量组在 4h内排泄量占总排出量的95 %以上。结论 :GN在病人体内无蓄积 ,是一个安全的药物

The CMNa blood concentration time data after single dose and multiple dose were simulated by computer software, and they fit linear open two departments model. The CMNa concentration time data, pharmacokinetic parameters and elimination rate in urine between single dose and multiple dose were compared. RESULTS CMNa distributed and eliminated rapidly in patients, their concentration time and pharmacokinetics parameters and elimination rate between single dose and multiple dose were similar with each other, and they have no significant difference. CONCLUSION CMNa will not accumulate in patients′ bodies, and it is a safe drug.
