Serial sections of the spinal cords of two adult animals were compared as to the configuration of the white and gray matters and to the relative magnitude of the motor cells in the anterior horn of the cervical and lumbar enlargements. The results were as follows: The cervical enlargement innervating the foreleg is much larger in cross section than the lumbar enlargement which innervates the hindleg. As compared to the lumbo-sacral levels, the anterior horn of the cervical region is broader in area and its cell columns attain greater proportions with better delimitation. In the cervical enlargement, the gray matter of the anterior horn expands laterally to form the antero-external and postero-external angles. The latter thrusts far out and contains the retroposterolateral column supplying the digital muscles. At equal-spaced representative levels, the cervical enlargement consis- tently out-ranks the lumbar enlargement in the distribution of the motor cells in the anterior horn, which control the leg muscles. The sum total of such cells at all the levels of the cervical enlargement is 1.4-1.5 times that of the lumbar enlargement. Moreover, when taken separately, the total motor cells in the posterior part of the anterior horn of the cervical enlargement, which innervates the distal portion of the foreleg, are 2.2-2.8 times that of the lumbar enlargement. The third and fourth toes assume enormous size on the forefoot. The retroposterolateral column which supplies the digital musculature is larger and longer in the cervical intumescence. The cells in the middle segment of this column are generally larger-sized, more numerous and with better-defined subdivisions. These traits apparently suggest that the cells in control of the third and fourth toes are mainly located at this plane. The striking difference in the size of the fore and hind feet reflects itself in the differential development of the anterior horn cells in the cervical and lumbar enlargements. This goes to show once more that the motor centers of the spinal cord develop in correlation with the peripheral organs innervated as well as with the particular mode of life of the animal.

Biochemical analysis of human placenta has showed that the purified LH-RHextracted from placenta after the 12th week of gestation is immunologically,physio-chemically and biologically indistinguishable from the hypothalamic principle(Gibbons et al 1975;Siler-Khodr et al 1978;Khodr et al 1980).In a recent report,Zhu and Zhang(1982)have provided evidences to show the presence of LH-RHin the human trophoblastic tissue at the age as early as Day 45 of pregnancy.Thefinding leads us to postulate that a regulatory role of this peptide may be involvedin the mediation of placenta function during pregnancy.An in vitro study of theeffect of a nona-peptide LH-RH([D-Ala~6-Des-Gly-NH_2~(10)]-LH-RH-ethylamide,LH-RH-A)on the incorporation of tritiated uridine,thymidine and leucine intohuman trophoblastic tissue was,therefore,carried out in the present investigation.In addition,the presence of LH-RH receptors in the trophoblastic tissue was alsoidentified and characterized.Human placentas between 6—8 week of gestation were collected at the timeof suction curettage.The villous tissue was carefully separated from the decidualtissue by taking good care to exclude the remnant of membranes.The villoustissues were then diced and pooled together,they were suspended in PBS(pH 7.4)and an aliquot of 0.2 ml of the suspention(equivalent to 20—30 mg)was addedto each of the incubation tubes.For the labelling of trophoblastic RNA,DNA andprotein,tritiated uridine,thymidine and leucine were added individually into eachof the incubation tubes,which were incubated for 90 minutes at 37℃ on a Dubnoffshaker with or without LH-RH-A.In some tubes,75 μg of actinomycin D orcyclohiximide was added.By the end of incubation,the suspensions in each tubewere washed three times with 3 ml PBS.They were then transferred into vialscontaining 0.5 ml of 1 N NaOH and left at 37℃ overnight.An aliquot of 20μlfrom each vial was taken for protein determination according to the method of Miyachi et al.The results show that the specific radioactivities of the LH-RH-A treated groupsat the dosages ranging from 1—100μg were significantly increased as comparedwith that of the control group.The presence of actinomycin D or cycloheximidein the medium remarkably inhibited RNA or protein synthesis.Analysis of the ratiosof the ~3H-and ~(14)C-leucine in each single band on the gel slabs showed that thepepfide had no specific stimulatory effect on the protein synthesis of the trophoblastictissue.It is of interest to note that there is a difference in physiological effect ofLH-RH-A on the decidual tissue and the trophoblastic cells,i.e.,inhibitory in theformer case and stimulative in the latter.The underlying mechanism of this para-doxical effect is unknown.The LH-RH receptors of the trophoblastic tissue were analysed by Scatchardplot which showed that the binding of ~(125)I-LH-RH-A to the trophoblastic tissue wassaturable,being a single class of binding sites with high affinity(K_α=4.8±0.6×10~9M~(-1)) and low capacity(50.2±4.4 fmol/mg protein).The finding of the present experiments provides an unequivocal proof toshow that LH-RH is directly involved not only in control of the secretory func-tion of trophoblast,but also in regulation of the metabolic activities of the placenta.
