Objective To investigate the possibility to fabricate bone graft in the shape of human mandibular ramus possessing blood vessel pedicel employing the method of tissue engineering. Methods Marrow stem cells (MSCs) were cultured, expanded and induced in vitro. Cells were seeeed into porous natural coral(NC) in the shape of human mandibular ramus complex at the density of 2×10 8 /ml in 200 ul cell suspension. After two days in vitro incubation, five cell coral complexes were implanted into cell donor rabbit under inferior epigastric artery to prefabricate vascularize bone graft of specific shape.Two months after operation, bone formation was observed by gross inspection, electronic microscope observation, and histological observation. Results Two months after operation, well vascularized bone grafts in the shape of the initial scaffold and with blood vessel pedicel were regenerated successfully. Osteogenesis process occued in the manner of endochondral bone formation.New bone formation could be observed by electronic microscope observation and histological observation on the surface and in the pores of NC. Conclusion Fabrication of vascularized bone graft in predetermined shape could be successfully achived employing the method of tissue engineering. This kind of bone graft may be very useful in future clinical application.

目的 观察用组织工程方法再造下颌骨升支 ,并使之含有知名血管蒂的可能性。方法 体外培养、扩增、诱导兔骨髓基质干细胞 (MSCs) ,将细胞接种于人下颌骨升支外形的多孔珊瑚中 ,体外培养 2d后 ,植入MSCs供体兔腹壁下动静脉深面 ,预构血管化的人下颌骨升支。术后 2月取材 ,通过大体标本观察、扫描电镜检查、组织学检查 ,观察骨组织的形成情况。结果 在兔体内再造出了腹壁下动静脉为蒂的骨组织 ,并具有人下颌骨升支的解剖形态 ;X线检查显示有X线阻射影 ,珊瑚有部分吸收 ;组织学检查结果证实 ,在NC表面及孔洞内均有新骨形成。结论 用组织工程方法可预构出具有特定形态的血管化的骨组织

目的构建钛网为支架增强的组织工程骨,修复下颌骨节段性缺损(25mm),观察下颌骨修复形成情况。方法羊骨髓基质干细胞转化的成骨细胞与高孔隙率滨珊瑚颗粒复合后,钛网为加强外支架,修复自体下颌骨缺损,行X线观察、大体及组织学观察下颌骨修复情况。结果X线观察实验组随时间延长,密度逐渐增加,大体观察见下颌骨下缘连续性恢复,组织学见支架中有大量新骨形成,并与钛网相融合。结论利用钛网加强的方法可以有效修复下颌骨节段性骨缺损。
