The incorporation rates of ~3H-TdR also decreased rapidly and the rates of cell proliferation changes to minus on 5th day. As for KB cells, it had an inhibitory effect when the concentration of EDF was 0.15μg/ml and it became obvious when its concenteation increased to 0.3μg/ml. All the results above shows that EDF has an inhibitory effect on the proliferation of tumor cells and the effect is cell type dependent as well as dose-dependent.

从兔网织红细胞提纯的红细胞分化调节因子(erythroid differentiation factor,EDF),能对体外培养的自发转化成纤维细胞系L929及人鼻咽癌细胞系KB产生作用。当EDF剂量为0.10μg/ml时,可引起L929细胞形态发生改变,并有细胞核固缩现象。第2d的细胞生长抑制率为64.86%,软琼脂集落形成率为0;第5d时细胞增殖为负值。~3H-TdR掺入率明显降低。EDF剂量为0.15μg/ml时,对KB细胞生长已有抑制作用。EDF剂量达0.30μg/ml时,生长抑制明显。以上结果证明了EDF对恶性细胞具有增殖抑制作用。这种作用对不同种类细胞敏感性不同,并且与剂量呈正相关。

The nasopharyngeal cell line CNE-2Z-H5,F1 and lymph node of H5 transplantable tu-mor and lung metastatic tissues were xenografted subcutaneously to node mice,The characteristics of spono-taneous metastases were observed.Furthermore,we have studied the differences in the doubling time,thetumor weight,and the expression of proliferating cell nuclear antigen(PCNA)between metastatic groupand nonmetastatic group at different time,The results showed thet the metastatic ability and pathways inall kinds of transplantable tumors were different.The tumor weight of metastatic group increased fasterthan nonmetastatic group(P<0.05),the doubling time of metastatic group was shorter than nonmetastaticgroup(P<0.05),and the expression of PCNA in metastatic group was stronger than that in nonmetastaticgroup within some times.These results suggested that there may be some heterogeneous subpopulation ofdifferent metastatic ability and pathways in lymph node and lung metastatic foci.There are some relation-ships between the metastasis and proliferation ability.

将鼻咽癌克隆细胞株CNE-2Z-H5、F1及H5移植瘤之淋巴结及肺转移灶组织,移植入裸鼠皮下,观察其自发性转移;同时研究了转移组和非转移组移植瘤之间,不同生长时间瘤重、体积倍增时间及增殖细胞核抗原表达的差异。结果显示:各类移植瘤的转移能力及途径均不同;在一定生长时间内,转移组移植瘤重大于非转移组(P<0.05).体积倍增时间较非转移组短(P<0.05),增殖细胞核抗原表达较非转移组强。提示:淋巴结及肺转移灶组织中可能存在高转移特性、且为不同转移途径的异质性转移亚群;肿瘤细胞转移和增殖能力存在一定的关系。
