This study reports the features of porcine coronary artery smooth muscle cells in the primary culture and subculture with light and electron microscopes for the first time in China. These cells that have been growing in culture for less than 5~7 days have a spindle shape with sausage-shaped nucleus containing 2~3 nucleoli. The cytoplasm is phase dense and filled with organells such as bundle of thin filaments,dense area or body, and many cytoplasmic cavernae or vesicles. Golgi complexes, smooth and rough...

This study reports the features of porcine coronary artery smooth muscle cells in the primary culture and subculture with light and electron microscopes for the first time in China. These cells that have been growing in culture for less than 5~7 days have a spindle shape with sausage-shaped nucleus containing 2~3 nucleoli. The cytoplasm is phase dense and filled with organells such as bundle of thin filaments,dense area or body, and many cytoplasmic cavernae or vesicles. Golgi complexes, smooth and rough ER, and mitochondria often completely encircle the nucleus. A defined basal lamina is the major distinctive character. The cultured cells tend to grow into series of "hill" and "valley". The trypsin-treated cells can be induced to contract by mechanical or cold stimulations. The above results show that cultured porcine coronary artery smooth muscle cells may remain their initial phenotype under concerned condition in vitro. Therefore,we believe that this system of culture cells is of great value in research for pathogenesis of atherosclerosis and its subsequent complication, as well as for the mechanism of coronary artery spasm in acute myocardial infarction.

本文首次于国内报道原代和继代培养猪冠状动脉平滑肌细胞的光、电镜结构特征。培养5~7天时细胞呈梭形,细胞核腊肠形,含2~3个核仁。胞浆相差大,充满细胞器,如微丝束、密区和密体、数量较多的质膜小凹或小泡.高尔基氏复合体、粗面内质网、平滑内质网和线粒体位于核周。基膜明显为其显著特征。细胞生长方式呈“峰谷”样。机械或冷刺激可诱导继代培养的细胞产生收缩。上述结果表明,在一定培养条件下猪冠状动脉平滑肌细胞在体外仍可保持其原有的表现型。因此,作者认为这个培养细胞系统对研究动脉粥样硬化及其并发症和急性心肌梗塞中冠状动脉痉挛的机理具有很大的应用价值。

Anoxia caused a transient relaxation lasting 4.3± s 0.8 min, followed by a sustained contraction in the isolated porcine coronary artery stimulated by KC1 25 mmol·L; the removal of the endothelium abolished the anoxia induced relaxation of the coronary artery, and almost eliminated the contraction response. Glibenclamide (Gli) 0.1, 0.5, 2.5 and methylthioninium chloride (MC) 5, 10 μmol·L-1 produced a concentration-dependent decrease in the relaxation response of the coronary artery caused by...

Glibenclamide (Gli) 0.1, 0.5, 2.5 and methylthioninium chloride (MC) 5, 10 μmol·L-1 produced a concentration-dependent decrease in the relaxation response of the coronary artery caused by anoxia, and a combination of Gli 2.5 and MC 10μmol·L-1 almost abolished the dilation. These data indicate that anoxia-induced transient relaxation of the isolated porcine coronary artery possibly is mediated by the combined effects of the ATP-sensitive potassium channel and the endothelium-derived relaxing factor (EDRF).

血管内皮细胞通过释放内皮细胞源性收缩和舒张因子调节血管平滑肌细胞的舒缩活动,经典上在离体血管实验中常采用乙酰胆碱作为检验血管内皮细胞完整性的工具药。最近我们在离体猪冠状动脉环上观察到不论内皮是否完整,乙酰胆碱均呈剂量依赖性收缩反应,但三磷酸腺苷则可诱发内皮依赖性松弛反应,在此基础上我们建立了以三磷酸腺苷为工具药评价猪冠状动脉内皮细胞完整性的新方法。
