Hsien and Keng subspecies of Asian cultivated rice (O. sativa L.) are originated from wild rice at same period, there are not the problem of fundamental type and variation type.

采用程侃声(1993)亚洲稻籼粳亚种六项形质性状鉴别方法,对普通野生稻(O.rufipogon)进行籼粳性状分化的调查研究,了解作为栽培稻近缘祖先种的普通野生稻各生态类型的籼粳性状分化情况。结果表明,普通野生稻各生态类型在叶毛、穗轴1、2节长、谷粒长宽比等性状上已有籼粳性状表现的差异,在同一性状中各生态类型群体上表现也有差异;有的性状(叶毛)在同一植株不同生育期也表现出籼粳的差异。因此认为普通野生稻存在着籼粳性状的结构基因,性状上的籼粳差异是基因表达调控不同的结果。亚洲栽培稻籼粳亚种可能同时起源于野生稻,不存在基本型与变异型问题。

Purpose:Study on tumor heterogeneity of Nasopharyngeal Carcinoma cell line SUNE 1.Methods:The potential of malignant transformation and metastatic ability,and the gene expression of four subclones(5 8F,9 4E,6 10B,13 9B)of SUNE 1 were studied by means of the ability colony formation in soft agar,animal experiment and RNA fingerprinting map.Results:(1)the heterogeneity exist in the potential of malignant transformation and the gene expression of four subclones.(2)The highest(5 8F)or lowest (13 9B)tumorigenic and metastatic subclones have been selected.(3)The heterogeneity of gene expression exist in four subclones and the different expressed fragments were selected.Conclusion:Our study suggested that four subclones that originated to SUNE 1 are different single colony cell lines.Four subclones may be useful experiment materials for cloning genes in relation with the development of NPC as well as provide experimental evidences for further research on gene therapy,chemotherapy and immunotherapy.

目的:对鼻咽癌细胞株SUNE1异质性进行研究;方法:应用软琼脂集落形成实验、动物实验及RNA指纹图谱差异显示对SUNE14亚株58F,94E,610B,139B进行体外转化能力、成瘤性、转移能力及基因表达差异进行研究。结果:①4亚株体外转化能力、成瘤性和转移能力存在差异;②筛选出高成瘤、高转移潜能的亚株58F、94E,只成瘤不转移的610B,不成瘤的139B;③4亚株的基因表达存在异质性,并筛选出3条迁泳率不一致的差异片段。结论:SUNE14亚株为起源于同一母系的不同的单克隆细胞亚株;为筛选和克隆NPC发生发展相关基因的研究提供实验材料,亦为NPC的基因治疗、免疫治疗及化学治疗的研究提供实验依据。

On the aspect of structure, all proteins coded by LIM class homeobox genes contain homeodomain and LIM domain; on aspect of biological function, all LIM class homeobox genes are related in the determination of embryonic cell lineage and differentiation of embryonic cells in early development. We began to work with LIM class homeobox gene in order to study the molecular mechanism of early development of amphioxus (Branchiostoma belcheri tsingtaoense).While LIM class homeobox gene Bblim in amphioxus was being cloned, an intron was found in the homeobox of this gene. Such a structure of this gene is different from that of most other LIM class homeobox genes.1. MATERIALS AND METHODS

LIM类同源框基因(homeoboxgene)保守性很强,在动物界广泛存在。各种LIM类同源框基因的共同点是:在结构上,LIM类同源框基因的编码蛋白都具有同源域和LIM结构域;在生物学功能方面,都与早期发育中胚胎细胞谱系的决定和胚胎细胞分化有关。本实验提取青岛文昌鱼基因组DNA,用一对简并引物对其LIM类同源框基因Bblim进行PCR扩增,将所得DNA片段克隆到pBluescriptⅡKS(+)质粒上,经测序发现,其同源框区比大多数LIM类基因的同源框区要长得多,其上插入有一个长度为138bp的内含子(Fig.1)。这个内含子的序列中有真核生物核基因mRNA内含子的典型序列。并且与该基因cDNA克隆的测序结果相吻合。从而,可以确认该序列中这个内含子的存在。已知绝大多数LIM类同源框基因的同源框区无内含子。因此,有必要对其作深入研究。
