The cellular effects of glucocorticoids on proliferation and differentiation of a human inegakaryoblastic leukemia cell line (HIMeg) were studied in comparison with sex steroid hormones. In both liquid and methylcellulose culture systems, glucocorticoids suppressed the proliferate of HIMeg cells in a dose-dependent manner. The cellular effects of glucocorticoids on proliferation and differentiation of a human inegakaryoblastic leukemia cell line (HIMeg) were studied in comparison with sex steroid hormones. In both liquid and methylcellulose culture systems, glucocorticoids suppressed the proliferate of HIMeg cells in a dose-dependent manner. In contrast, sex steroid hormones except progesterone had little effects on the proliferation of HIMeg cells. In liquid culture systems, morphological changes were not evident, and the percentage of cells with multilobular nuclei increased slightly from 2% to 8% after glucocorticoid treatment. Similarly, only 2% of HIMeg cells expressed glycoprotein Ⅱb/Ⅲa (GP Ⅱb/Ⅲa) antigen without hormone, whereas 30% of the cells expressed GP Ⅱb/Ⅲa with the addition of glucocorticoids. These data indicated that glucocorticoids could induce differentiation of HIMeg cells. To clarify the molecular mechanisms, glucocorticoid receptor (GR) expression was examind by Scatchard analysis, and it was found that there was a saturable, high affinity GR in HIMeg cells. Furthermore, the cellular effects of glucocorticoids on HIMeg cells could be reversed by RU486, a potent glucocorticoid antagonist. These observations suggest that the cellular effects of glucocorticoids on HIMeg cells were mediated by GR.

Our recent studies showed that glucocorticoids play an important role in the processes responsible for proliferation and differentiation of a human megakaryoblastic leukemia cell line (HIMeg) and that the effects of glucocorticoids are mediated by glucocorticoid receptor(GR) in these cells.The regulation of GR expression in HIMeg cells by retinoic acid was further studied utilizing radioligand binding assay and molecular hybridization analysis.It was shown that retinoic acid could significantly increase the GR binding activities in a time-dependent manner,reaching a maximum of about 2-fold 24h following treatment.Dot blot hybridization analysis utilizing 32P-labeled GR cDNA fragment as a probe showed that GR mRNA was also up-regulated by retinoic acid,suggesting that the regulating effects of retinoic acid on GR occurred at least partially at GR mRNA levels. To explore further the possible consequence of retinoic acid-induced regulation of GR exp ression in HIMeg cells,the combined effects of glucocorticoids and retinoic acid on HIMeg cell proliferation were evaluated.It was found that the inhibitory effects of combinations of glucocorticoids and retinoic acid on HIMeg cell proliferation showed an additive property,suggesting that the regulation of GR expression by retinoic acid was of biological significance.

Objective:The purpose of this paper was to study the effect of recombinant human gamma interferon(rhIFN-γ) on glucocorticoid receptor(GR) expression in HOS-8603.cells,a human osteogenic sarcoma cell line. Methods:The GR binding activity and GR mRNA levels in HOS-8603 cells treated with or without rhIFN-γ were determined by radioligand binding assay and molecular hybridization analysis,respectively.Results : Treatment of HOS-8603 cells with rhIFN-γ resulted in down-regulation of GR number , with no change in the binding affinity for glucocorticoids.The maximum decrease in receptor binding was evident at 10 IU/ml rhIFN-γ concentration.Time-course studies revealed that the effect reached a maximum at 36 h treatments. To clarify the molecular basis for the down-regulation of GR by rhIFN-γ,change in GR mRNA levels was further investigated by RNA blot hybridization analysis.It was found that there also existed a time-dependent decrease in GR mRNA levels in HOS-8603 cells after treatment with rhIFN-γ.In the presence of rhIFN-γ,the inhibitory effect of glucocorticoids on HOS-8603 cell proliferation was blunted.Moreover,the induction of alkaline phosphatase activity by glucocorticoids was attenuated in response to rhIFN-γ treatment.Conclusion:The results of this study suggest that rhIFN-γ may influence GR activity which at least partially occurs at mRNA levels,and that the decrease in receptor activity in HOS-8603 cells parallels with the decrease in glucocorticoid-mediated functional responses.

以人骨肉瘤细胞系(HOS-8603)为模型,研究重组人γ干扰素(rhIFN-γ)对糖皮质激素受体(GR)的调节作用及其生物学意义。方法:GR结合活性的测定采用放射配体结合分析法;GRmRNA的测定用分子杂交法。结果:极低浓度的rhIFN-γ对GR具有下调作用,但对受体亲和力无明显影响;量效关系研究表明,当rhIFN-γ浓度为10IU/ml时,GR浓度已降至最低水平;时效关系研究表明,rhIFN-γ作用36h后对GR的下调作用达到最大效应。用分子杂交方法研究表明,rhIFNγ下调GR的同时,伴有GRmRNA水平的相应降低;rhIFN-γ本身虽然对HOS-8603细胞的增殖分化过程无明显影响,但却可明显削弱糖皮质激素对该细胞系的增殖抑制作用和对碱性磷酸酶活性的诱导作用。结论:rhIFN-γ对HOS-8603细胞GR有非常明显的下调作用,且主要由GRmRNA表达减弱引起;rhIFN-γ对GR的调节作用伴有受体功能活性的相应降低,表现为细胞对糖皮质激素的反应性明显减弱。
