With this assay 4 tri-halogenated anilines and 1 tri-halogenated benzene were tested for their ability to generate CO during microsomal metabolism. Among those compounds tested 2,4,5-trifluoroaniline was the only one able to generate significant amount of CO.Different from the CO generation specificity the de-fluoridation rates of 2,4,5-or 2,4,6-trifluoroaniline were similar. Phenobarbital or Dexamethasone pretreatment on rats increased the CO generation from 2,4,5-trifluoroaniline and showed little influence on other compounds tested.These results revealed that the metabolic CO generation from halogenated anilines was specific for substrate.
