Human immunodeficiency virus (HIV), an etiologic agent of AIDS, is a retrovirus. Its protease cleaves the primary translational products of the genome-size mRNA to make the virus replica normal. It is difficult to further study and apply HIV-1 Pr for there have been no satisfied system of expression and activity detecting. In order to further study HIV Pr, we first amplified whole gene of the HIV-1 Pr from a cDNA clone of a HIV-1 strain ARV-2 by polymerase chain reaction(PCR),meanwhile we also introduced the restriction enzyme and HIV pr recognition sites into the Pr gene. To express HIV-1 Pr, the Pr gene, was re-inserted into prokaryotic expressing vector pSM43, and the recombinant expressing plasmid pSPX414 were cultured at 32℃ and induced at 42℃ for 4 hours until reaching the best expression. The expressed Pr accounts for 30.5% of the total bacterial proteins. By Western-Blot analysis, we found that HIV-1 postive antiserum and antiserum to HIV-1 Pr could combine recombiant HIV-1 Pr well, and the activity of the expressed HIV-1 Pr was good enough also.

艾滋病是本世纪80年代初发现的一种烈性传染病,5年病死率为100%,致病因子为人免疫缺陷病毒,该病毒的蛋白酶在病毒复制和成熟中具有决定性的意义。由于目前国内外尚未获得艾滋病病毒蛋白酶的高效表达的重组子及活性检测系统,限制了它的研究与应用。本文利用PCR技术修饰了艾滋病病毒蛋白酶的基因,使其具有便于克隆及表达用的限制酶切位点及转录终止码,井在其C末端设置了一个可用于检验该酶活性的特殊序列。DNA序列分析揭示上述突变策略成功,将修饰后的艾滋病病毒蛋白酶基因克隆入大肠杆菌表达系统,并获得高效表达(>30%),Western-Bolt鉴定结果表明所表达的蛋白为艾滋病病毒所特有,并具有较好的生物活性。

The recombinant was identified with restrictive digestion and sequencing methods, and then was transformed into 293 cells with liposome, the expression of P1 A in 293 cell transformed with the recombinant was checked by ECL western-blot and immunohistochemistry. Results The recombinant P1A/ pSMART plasmid had been constructed correctly and could express the target P1A gene in transformed 293 cells correctly. Conclusion We had constructed successfully the recombinant P1A/pSMART plasmids which could be used as tumor vaccine for inoculatation into tumor animal model at next step and the immunological effect can be determined further.

目的 克隆小鼠肥大细胞瘤P815细胞株的肿瘤特异P1A基因于含α-病毒复制酶的真核表达载体pSMART中,以制备P815肿瘤DNA疫苗。方法RT-PCR方法扩增P1A基因,以含α-病毒复制酶的哺乳细胞高效表达质粒pSMART为载体,构建重组肿瘤DNA疫苗。重组体经酶切和测序后,再用脂质体转化293人胚肾细胞,ECL western-blot法和免疫组化法鉴定转化细胞中P1A基因的表达。结果 正确构建了P1A/pSMART重组质粒,并且在转化此质粒的293细胞中检测出了P1A的表达。结论 成功构建了重组P1A/pSMART肿瘤DNA疫苗,可以进行下一步的肿瘤动物模型的疫苗接种及疗效观察。
