Results: Primary cultured RPE cells contained large amount of pigments in polygonal shape. Passages of cultured RPE cells presented transparent spindle shape. Electron microscopy demonstrated that the normal morphological characteristics of rabbit RPE in vivo were maintained in cultured cells, which included cell polarity, pigments, intracellular organelles, and cell junction complex. Conclusion: This technique allowed us to produce a large quantity of RPE cells for in vitro experiments.

Objective:To investigate the effect of the high glucose concentration on the intercellular adherens junction complex in the cultured human peritoneal mesothelial cells(HPMC),and clarify the cause of decreased dialysis efficiency during prolonged peritoneal dialysis. Methodology:All test were performed on the human peritoneal mesothelial cell line(HMrSV 5).Three groups were divided.They were ①normal glucose group treated with 5.5 mmol/L D-glucose;②high glucose group treated with 140 mmol/L D-glucose;and ③high osmolality group with 5.5 mmol/L D-glucose and 140 mmol/L mannitol.Cell detachment and viability were observed at the time of 6,12,24,and 48 hours.The mRNA expression of E-Cadherin and β-Catenin was detected by semi quantitative reverse transcription polymerase chain reaction (RT-PCR),and the protein measured by Westernblot and immunofluorescence.High glucose-induced actin disassembly was determined by fluorescence labeling technique. Results:High glucose caused a time-dependent increase in the cell detachment.The mRNA expression and level of intercellular adherens junction protein(E-Cadherin and β-Catenin)were decreased,and the fluorescence expression immunostaining by anti-E-Cadherin and anti-β-Catenin antibodies was weaker and often discontinuous along the cell contour.When HPMC was incubated in high glucose,F-actin bundles appeared less prominent and changed to irregular network.Mannitol had no effect on actin assembly and on the expression of E-Cadherin and β-Catenin at the same osmolality. Conclusion:These findings suggest that the peritoneal mesothelium is damaged by prolonged peritoneal dialysis using high glucose dialysate,and that impairment of the intercellular adherens junction complex of HPMC by high glucose play an important role in cell detachment and a progressive reduction in dialysis efficiency.

To discuss the mechanism of dexamethaxone on cytoskeletal reorganization of three - dimensional rats hepatocyte spheroids. Rats hepatocytes were cultured on primary dishes for spheroids formation. The hepatocytes spheroids were again cultured with type I - C collagen matrix in Willam' s E medium, in which dexamethasone were added, then observed by phase -contract microscopy. The cytoskeletal reorganization was observed by immunocytochemical stain with Z0-1. The levels of proteins expression, such as junction complex proteins, ERM proteins, small GTPase protein and Pgp, were detected by Western blot. Tight junction protein Z0-1 strongly accumulated in the walls of cysts treated with dexamethasone. The levels of proteins, such as junction complex, Pgp, rho A and radixin, were incereased after treated with dexamethasone. The levels of rho A and radixin were corresponded with the rate of cyst formation. Dexamethasone can increase bile secretion, improve the levels of rho A and radixin, induce accumulation of actin and junction complex proteins in the walls of the cysts, and induce cytoskeletal reorganization and cysts formation. The three- dimensional culture system will be useful for hepatocytes research.

探讨地塞米松对大鼠肝细胞球状体细胞骨架再建的影响机制。肝细胞球状体培养于胶原三维基质中,行ZO-1免疫组化染色;抽提蛋白行蛋白质的印记杂交。ZO-1在肝细胞球状体空泡壁聚集,地塞米松促进Pgp及细胞粘连相关蛋白质的表达,促进小GTPase超家族蛋白Rho A和ERM超家族radixin的表达,Rho A、radixin表达水平与空泡形成率相一致。地塞米松促进大鼠三维培养的肝细胞胆汁分泌,在Rho A和radixin介导下肌动蛋白重新分布,细胞骨架再建,调节紧密连接功能和聚集,导致空泡样结构的形成。
