Using a polymerase chain reaction based telomerase activity assay (TRAP),we examined telomerase activity in 5 normal bladder tissues, 4 bladder infection tissues, 42 bladder cancer tissues and 42 adjacent noncancerous bladder tissues. None of the normal and inflammatory bladder tissue exhibited telomerase activity. 32 of the 42(76.19%) bladder cancer tissues revealed telomerase activity. None of the normal and inflammatory bladder tissue exhibited telomerase activity. 32 of the 42(76.19%) bladder cancer tissues revealed telomerase activity. In the adjacent normal tissue taken from the bladder cancer, 11 of the 42(26.19%) were telomerase positive. The telomerase is more frequently activated in advance ,not well differentiation bladder tumors. The results indicate: telomerase activation has strong relationship with the invasion and development of bladder cancer. The telomerase activation of the paracancer tissue shows the possibility of microinvasion.

应用聚合酶链反应(PCR)为基础的端粒酶活性检测方法(TRAP法),对5例正常膀胱组织、4例膀胱炎症组织、42例膀胱癌组织和癌旁正常组织进行检测。正常膀胱组织和膀胱炎症组织无端粒酶活性,膀胱癌组织端粒酶表达阳性率76.2%,癌旁组织阳性率为26.2%,端粒酶活化与膀胱癌分期分级呈正相关。结果提示:端粒酶活化与膀胱癌浸润发展密切相关,癌旁组织端粒酶活化提示肿瘤微浸润可能。

Objective: To explore the influence on telomerase activity of bladder cancers after the treatment

目的:研究卡介苗细胞壁骨架(BCGCWS)加白细胞介素2(IL2)膀胱灌注对肿瘤组织端粒酶活性的影响,从而探讨该治疗的作用机理。方法:采用端粒重复序列扩增法(TRAP)分别检测20例膀胱肿瘤用药前后端粒酶活性的表达。结果:经分别测定端粒酶提取原液以及10倍、100倍稀释液中端粒酶活性的表达,表明用药后肿瘤组织中端粒酶的活性明显降低(P<001)。结论:BCGCWS联合IL2膀胱灌注能有效地抑制肿瘤细胞端粒酶的活性,提示该治疗的作用机理之一可能是肿瘤端粒酶活性的改变。

Objective To study the telomerase activity in exfoliated urothelial cells of bladder cancer and to evaluate its clinical significance. Methods Telomerase activity was studied by means of a modified PCR based telomeric repeat amplification (TRAP) silver stain assay in tissue and urine samples and bladder washings from 48 cases of bladder cancer and 12 normal bladder and in urine samples and bladder washings from 20 patients with non urothelial cancer. Results Telomerase activity was detected in 44 of the 48 tumor samples (91.7%) while none of the normal bladder tissue showed telomerase activity.Telomerase activity was also detected in 83.3% (40/48) of urine samples and 87.5%(42/48) of bladder washings from bladder cancer patients.In 12 patients with well diffentiated bladder cancer (G 1),telomerase activity was detected in 9 urine samples (75.0%) and in 10 bladder washings (83.3%). Conclusions Telomerase activity could be detected in most urine samples of bladder cancer patients and therefore would be a good diagnostic marker.But it is not related to the tumor grade or stage.

目的检测尿脱落细胞端粒酶活性并探讨其临床意义。方法应用改良的端粒重复序列扩增(TRAP)银染方法,分别对膀胱癌组织、正常膀胱组织,以及膀胱癌患者和非尿路上皮肿瘤患者的尿脱落细胞、膀胱冲洗液进行端粒酶活性检测。结果12例正常膀胱组织均无端粒酶活性,48例膀胱癌组织中44例(91.7%)端粒酶阳性。膀胱癌患者尿液及膀胱冲洗液中脱落细胞端粒酶阳性率分别为83.3%(40/48)和87.5%(42/48)。12例分化良好(G1级)膀胱癌患者中,尿液和膀胱冲洗液中脱落细胞端粒酶阳性率分别为75.0%(9/12)和83.3%(10/12)。结论尿脱落细胞端粒酶活性检测敏感性高,可用于膀胱癌的早期诊断和术后随访。
