    Inbred Spraque-Dawly female rats were hypophysectomized by the sterostactic approach through eardrum. Donor spleen cells (SC) were isolated from inbred newborn Wistar rats and activated in vitro with ConA. The donor specific tolerance of the hypophysectomized rats was induced by, (1) injecting different doses of the activated spleen cells (ASC) intravenously into the recipient rats (Hpox-SD), (2) injecting cyclophosphamide (CP) intraperitoneally, (3) combined injections of ASC and CP. The donor specific tolerance of the hypophysectomized rats was induced by, (1) injecting different doses of the activated spleen cells (ASC) intravenously into the recipient rats (Hpox-SD), (2) injecting cyclophosphamide (CP) intraperitoneally, (3) combined injections of ASC and CP. The result showed that the single injection of ASC or the combined injections of ASC and CP could prolong the skin allograft survival times with a mean survival time (MST) being more than 33. 5 and more than 47. 8 days respectively. The above result indicates that ASC or ASC plus CP could successfully induce the donor-specific tolerance in hypophysectomized rats.

CsA gave a noticeable synergistic effect to the induction of immunological tolerance. MLR and CD 8 + T cell were remarkablely reduced( P <0 05) in the cases of component transfusion. This study suggested that components transfusion may be of clinical use for donor specific tolerance inducing.

    为探索诱导免疫耐受的途径和方法,作者采用大鼠异位心脏(颈部)移植模型,经外周途径输注不同品系供者的不同特异性抗原,包括供体特异性输血( D S T)、供体特异性脾细胞( D S S L)、供体特异性骨髓细胞( D S B M)。结果显示: D S T、 D S S L和 D S B M 均能诱导受者对移植物产生特异性免疫耐受,其中以脾细胞和骨髓细胞效果最好,平均移植物存活时间分别达到 429±667 和 403±116 天。混合淋巴细胞反应( M L R)和 T 亚群( C D8+ )检测在一定程度上反映了机体 T 细胞免疫状态,有助于对耐受的评价。成分输注供者特异性抗原作为刺激抗原,可望成为较理想的临床耐受诱导途径。
