The induction of donor - specific tolerance would eliminate the risk of long -term immunosuppression while ensuring allograft function and survival. Male SD rats were exposed to donor alloantigen by an intrathymic, intrasplenic, subeutaneous or intravenous injection of 2.5 ×107 male Wistar splenocytes, and 21 days later received a heterotopic Wistar heart transplant. The induction of donor - specific tolerance would eliminate the risk of long -term immunosuppression while ensuring allograft function and survival. Male SD rats were exposed to donor alloantigen by an intrathymic, intrasplenic, subeutaneous or intravenous injection of 2.5 ×107 male Wistar splenocytes, and 21 days later received a heterotopic Wistar heart transplant. Intrathymic injection of spleen cells combined with short course of CsA treatmenl could significantly prolong heart allograft. Our results confirmed the role of the thymus as a special site for the induction and maintenance of specific immunological unresponsiveness to organ allografts. T cell clonal deletion may play an important role in this immune tolerance.

本实验采用大鼠腹部心脏移植模型,在移植前21天将供体脾细胞分别注射到受体的胸腺、脾脏、静脉、皮下,然后行异位心脏移植,术后1周每日腹腔注射环孢素A(CsA)10mg/kg。胸腺注射供体脾细胞联合应用CsA组移植心平均存活时间较其它组显著延长,受体对供体靶细胞没有出现细胞介导的细胞毒作用.结果表明:胸腺注射洪体脾细胞联合应用CsA能成功诱导免疫耐受,胸腺内特异性T细胞克隆消除可能与免疫耐受的形式有关.

为了降低移植物免疫原性,诱导免疫耐受,本实验采用单抗加补体方法消除大鼠甲状旁腺移植物中的主要组织相容性Ⅰ类抗原(Ⅰa)阳性细胞,将其移植到同种受体肾包膜下,观察移植物平均存活时间。结果显示:处理组移植物平均存活时间为60天,对照组为14天,两者相比有极显著性差异(P<0.01)。说明消除Ⅰa抗原阳性细胞可显著延长大鼠甲状旁腺同种异体移植物存活时间。

Objective: Using donor skin sensitized recipients and illtraabdominal heterotopic cardiac transplantprocedure, we investigated the effect of donor pretreatment with tripterygium polyglycoside andrecipient pretreatment with intrathymic inoculation of donor splenocytes and their synergistic effectson the cardiac allograft survival. Methods: SD rat used as skin and heart donor and Wistar rat used asrecipient. All recipiets were sensitized with donor skin transplantation before heart transplantation(HT). There was single HT only in group A and donor pretreated HT in group B. After allogenicsplenocytes being inoculated in group C the heart was transplanted. In group D the two pretreat-ments were combined. ResuIts: The mean survival time ofcardiac allografts was much prolonged ingroup B, C and D in comparing with those in control group A. Microscopic examination showedthat the cell mediated rejection was much alleviated in group C and D, but the vascuIar rejection wasaggravated. Conclusion: Both of the method could evidently promote survival of cardiac allografts andact synergistically. Tolerance induction could obviosly depress the cell mediated immunity, but seemedto show no effect on prevention of the vessels injury caused by acute vascular rejection.

目的:用供体品系大鼠皮肤致敏方法及腹腔内异位心脏移植模型研究雷公藤多式供体预处理及供体脾细胞受体胸腹内注射诱导免疫耐受对移植心脏存活影响及二种方法的协同效果。方法:SD大鼠作供体,Wistar大鼠作受体,所有受体在心脏移植前均无用供体皮肤致敏,A组作单纯心脏移植,B组作经供体预处理后的供心移植,C组用供体脾细胞胸腺内注射诱导耐受后作心脏移植,D组考察供受体两种预处理方法的协同效果。结果:各处理组供心存活时间均校对照组明显延长,C,D二组病检见淋巴细胞浸润及心肌坏死明显减轻,但微血管病变较严重。结论:两种方法均能明显延长移植供心成活时间,并有良好的协同效果,经胸腺途径诱导的耐受能明显抑制细胞免疫,但对急性血管排斥没有预防作用。
