To investigate the gene expression of growth factor Wnt 5A in different phases during cell cycle at transcriptional level.To synchronize the renal cell carcinoma GRC 1 cell line was by double thymidine blocks and high pressure N2O gas methods . Using Semi quantitative RT PCR (reverse transcriptase polymerase chain reaction) to amplify the cDNAs of Wnt 5A in different phases. Result showed that the mRNA expression of growth factor Wnt 5A was detected in RCC GRC 1cell line. In S phase, the highest level of Wnt 5A was observed, medial and lowest was in G1 and M phase, respectively. The differences between S and M stages were significant ( P <0 05).Therefore growth factor Wnt 5A has the potential effect on tumorigenesis, It contributes to all phases during cell cycle, but gives more influence to S phase during cell cycle.

目的 研究转人 IL - 2基因对原代肾细胞癌 (RCC)基因表达的影响 ,为进一步对 RCC的基因治疗打下基础。方法 将人 IL - 2基因与逆转录病毒载体 p L NCX连接 ,转染包装细胞 ,经鉴定后 ,感染原代 RCC细胞 ,EL ISA法检测转人 IL - 2基因的原代 RCC的 IL - 2的表达 ,再以免疫组化法检测转人 IL - 2基因的原代 RCC和未转染原代 RCC的癌基因表达情况。结果 转人 IL - 2的 RCC细胞上清 IL - 2的浓度平均为 (390 8± 42 1) pg/ m l,人原代RCC细胞癌基因表达分析显示 EGFR、p185 erb B- 2、c- met和 bcl- 2均有轻度至中度的表达 ,转人 IL - 2基因的 RCC细胞的 p185 erb B- 2和 c- met的表达有一定程度的降低 ,而 EGFR和 bcl- 2的表达均没有影响。结论 转人 IL - 2基因可使原代 RCC的 p185 erb B- 2和 c- met的表达降低 ,为转人 IL - 2基因治疗肾细胞癌提供了部分理论基础。

目的研究肿瘤细胞表面HLA分子表达及IFN-γ的调控作用。检测TIL细胞中T细胞亚群的变化,并与PBMC比较。方法采用FACS检测细胞表面的CD抗原,采用ABC免疫组化染色检测肿瘤细胞HLA-I、II类分子的表达。以IFN-γ诱导建系的卵巢癌细胞表达HLA分子。结果肾细胞癌患者TIL细胞中的T细胞明显多于大肠癌患者。肿瘤患者PBMC中的T细胞数少于正常对照;肿瘤组织HLA-I类抗原的表达明显低于正常组织,HLA-II类抗原的表达高于正常组织。与正常组织相比较,大肠癌患者比肾细胞癌患者HLA-I类抗原的表达更低。IFN-γ可诱导建系卵巢癌细胞表达HLA-I类抗原,但不能诱导HLA-II类抗原的表达。结论肿瘤组织HLA表达的变化与肿瘤组织中TIL的数有关;IFN-γ能选择性地诱导卵巢癌细胞表达HLA-I类分子,在抗肿瘤免疫中具有重要的意义。
