Wheat seedlings were used as experimental material for investigating mechanisms of retardation of the leaf senescence by 6-benzylaminopurine (6-BA) in detached and intact leaves. It was shown that the leaves floating on water lost their chlorophyll but those supplied with 6-BA retained their green color for a period of 15 days. The free amino acid contents increase rapidly during the first 1-2 days after excision from the plants in both control and 6-BA-treated leaves, but a much lesser extent in the latter....

Wheat seedlings were used as experimental material for investigating mechanisms of retardation of the leaf senescence by 6-benzylaminopurine (6-BA) in detached and intact leaves. It was shown that the leaves floating on water lost their chlorophyll but those supplied with 6-BA retained their green color for a period of 15 days. The free amino acid contents increase rapidly during the first 1-2 days after excision from the plants in both control and 6-BA-treated leaves, but a much lesser extent in the latter. Similar results were obtained with leaves from nitrogen starved plants.None of the inhibitors (chloramphenicol, 2-thiouracil and 8-azaguanine) of nucleic acid and protein synthesis used counteracts the suppressing action of 6-BA.The respiration intensity was about the same in 6-BA-treated and control leaves, although leaf excision was followed by a strong rise in respiratory rate. There was no significant difference in the pattern of incorporation of ~(14)C—glucose into amino acids, organic acids and proteins between 6-BA-treated and control leaves and it is suggested that the suppressing effect on free amino acid accumulation in the detached leaves was due neither to the inhibition of amino acid formation from glucose nor to the stimulation of protein synthesis, but mainly to the prevention of protein degradation. In comparing the radiochromatograms of the free amino acids synthesized from the ~(14)C—glucose or ~(14)CO_2 in water and 6-BA-treated leaves. it was found that there exists a similarity between them in support of the above interpretation.In intact leaves, the application of 6-BA was effective in reducing free amino acid accumulation when the wheat seedlings were shaded with black cloth, whereas no effect was found under natural light.The difference of the responses may result from the shifting of the ratio between protein synthesis and breakdown in favour of the latter under artificial shading of intact wheat seedlings.A variety of evidences lead to the conclusion that retardation of leaf senescence by cytokinin is due mainly to prevention of protein breakdown, rather than to the promotion of protein synthesis.

Premature senescence, one of the major symptoms caused by waterlogging, could be quantitatively evaluated by the degradation of chlorophyll (chl) and the accumulation of malondialdehyde (MDA) in leaves of waterlogged wheat plants. The immediate precursor of ethylene, 1-aminocyclopropane-1-carboxylic acid (ACC), applied as a foliar spray to the seedlings during different periods enhanced the production of ethylene in wheat plants. Premature senescence, one of the major symptoms caused by waterlogging, could be quantitatively evaluated by the degradation of chlorophyll (chl) and the accumulation of malondialdehyde (MDA) in leaves of waterlogged wheat plants. The immediate precursor of ethylene, 1-aminocyclopropane-1-carboxylic acid (ACC), applied as a foliar spray to the seedlings during different periods enhanced the production of ethylene in wheat plants. In plants treated with ACC 10 days prior to waterlogging, chl destruction and MDA accumulation were much slower than in plants waterlogged only, this difference indicating the delayed senescence in the ACC-treated plants. By comparison, ACC applied 5 days after the onset of waterlogging promoted the destruction of chl and the accumulation of MDA, but ACC application to non-waterlogged plants showed neither chl destruction nor MDA accumulation. Moreover, Ethrel supplied at different periods of waterlogging acts as ACC. Experimental data presented support the assumuption that ethylene seems to be not a "trigger", but a promotor, in the acceleration of senescence of wheat plants; therefore, the decrease of ethylene production at the later stage during waterlogging is an adaptative process in response to waterlogging in the plants.

加速衰老——渍害的主要表现,可以叶绿素含量的下降和膜脂过氧化产物丙二醛含量的增加作为定量指标。在不同时期对小麦地上部喷施乙烯生物合成前体1-氨基环丙烷-1-羧酸(ACC)或乙烯发生剂乙烯利以增加体内乙烯产生,发现渍水前10天喷施,使小麦生长受一定程度抑制,可显著提高抗渍性。在渍水前5天或渍水同时给予ACC,植株的叶绿素和丙二醛含量与未处理的受渍植株的含量无显著差异,而渍水5天再进行喷施时,则进一步加速叶绿素分解和丙二醛的积累,加重了渍害。此时期以ACC或乙烯利处理不渍水的对照植株,既不加速叶绿素分解,也不促进丙二醛累积,表明乙烯具有加速衰老的作用,但不是渍水启动衰老加速的内因。

扫描电镜见正常早代体外培养二倍体细胞 SL_7细胞膜表面有许多微绒毛,在晚代则明显减少,代之而起是许多球状突起、细胞碎屑、赘生物,加入 CH_3的 SL_7晚代细胞膜表面微绒毛仍多而球状突起少。透射电镜见正常早、中、晚代细胞的有嵴线粒体、粗面内质网、溶酶体、空泡小体和胞核的变化,与文献报道 WI-38株同类细胞变化相似。随胞龄增加,有嵴线粒体、粗面内质网渐减,溶酶体、空泡小体变大、增多。CH_3组晚代 SL_7细胞的有嵴线粒体和粗面内质网较正常对照组细胞为多,溶酶体、空泡小体较少,它提示 CH_3有延缓细胞微细结构衰退和延缓细胞衰老的作用,为 CH_3临床效用提供分子细胞学的依据。
