s: To observe the protective function of well acupoints blood-letting method against acute ischemic brain injury. Rats were divided into sham operation group,cerebral ischemia group and treatment group. Acute ischemic brain injury was made by middle cerebral artery obstruction(MCAO). Well acupoints blood-letting therapy ,one drip each time ,was given in rats in treatment group immediately after cerebral ischemia. 1,3,6 ,24 hours after ischemia,rats were killed and c-fos protein(FOS)and HSP70 immune positive cells of cortex tissue in ischemic region were determined by immunohistochemistry assay to observe the influence on anti-apoptosis gene expression at early stages of acute ischemic brain injury. Increased expression of c-fos protein and HSP70 protein in cortex tissue of ischemic region was observed with no expression of HSP70 1 hour after ischemia,while the expression of FOS was increased. Expression of FOS and HSP70 tended to increase with the time elapsed during the following 3,6,24 hours. The count of c-fos and HSP70 immune positive cells in treatment group was all higher than that in model group during the same period of time with statistical significance.[Conclusion] Well acupoints blood-letting therapy can obviously enhance the expression of c-fos and HSP70 protein in ischemic region,increase the ability against apoptosis and the tolerance and adaptation ability of nerve cell to hypoxia and ischemia ,and enhance the neuron compliance after ischemia,thus further influencing the expression of objective gene at late stages,resisting the development of apoptosis,enhancing the repairing ability of brain. It is suggested that well acupoints blood-letting therapy has certain protective action against early stroke.

[目的]观察井穴放血法对急性缺血性脑损伤的脑保护作用。[方法]将实验大鼠分为假手术组、脑缺血组和脑缺血 +井穴放血治疗组 ,一侧大脑中动脉阻塞致急性缺血性脑损伤模型 (MCAO) ,治疗组在脑缺血后即刻给予井穴放血治疗 ,出血量为每穴1滴。分别于缺血后的1、3、6、24h处死 ,进行免疫组织化学染色 ,检测缺血区皮层脑组织c-fos蛋白 (FOS)及HSP70蛋白免疫阳性细胞 ,以观察井穴放血法对急性缺血性脑损伤后早期抑凋亡基因表达的影响。[结果]c-fos蛋白和HSP70蛋白在缺血区皮层脑组织表达增加 ,缺血后1hHSP70无表达 ,FOS表达增加 ,在随后观察的3、6、24h时程内HSP70和FOS均有随缺血时间延长表达增多的趋势。井穴放血治疗组在以上各时程c-fos及HSP70免疫阳性细胞数均高于同时段的缺血病模组 ,有统计学意义。[结论]急性脑缺血后应用井穴放血干预治疗可明显增强缺血区脑组织对抗神经细胞凋亡的即刻早期基因c-fos蛋白和抗应激HSP70蛋白的表达 ,从而提高了缺血后神经细胞对缺血、缺氧的耐受和适应能力 ,提高了缺血后神经元的可塑性 ,进而可影响晚期目的基因的表达 ,抵抗细胞...

Objective To investigate the preventive effect of blood-letting puncture of twelve Jing-Well points on acute ischemic cerebral lesion from intracerebral c-fos protein expression. Methods A rat model of acute ischemic cerebral lesion was made by coagulation occlusion of unilateral middle cerebral artery. Cerebral c-fos positive cell expression in the ischemic cortex was observed by immunohistochemical method and the preventive effect of blood-letting puncture of twelve Jing-Well points on the lesion was investigated....

Objective To investigate the preventive effect of blood-letting puncture of twelve Jing-Well points on acute ischemic cerebral lesion from intracerebral c-fos protein expression. Methods A rat model of acute ischemic cerebral lesion was made by coagulation occlusion of unilateral middle cerebral artery. Cerebral c-fos positive cell expression in the ischemic cortex was observed by immunohistochemical method and the preventive effect of blood-letting puncture of twelve Jing-Well points on the lesion was investigated. Results Acute ischemic cerebral lesion could quickly induce cerebral c-fos protein expression in the ischemic cortex, which tended to increase gradually with the prolongation of ischemic time. Blood-letting puncture of Jing-Well points as an intervention immediately after ischemia could significantly up-regulate cerebral level of c-fos protein expression in the ischemic cortex at 1,3,6 and 24 hrs after the ischemia. Conclusion C-fos protein expression is not only related to the severity of ischemia but also reflect the responsive ability of neurons. Cerebral ischemia can induce gradual expression of c-fos protein, suggesting that cerebral ischemic lesion can improve the ability of neurons to tolerate and adapt to ischemia and anoxia by a stress reaction and that early blood-letting puncture of Jing-Well points can further increase c-fos protein expression and improve the stress ability of neurons to ameliorate the ability of ischemic brain tissues to prevent the lesion and make repairs, slow neuronal apoptosis, prevent the further development of ischemic cerebral lesion and produce an effective protecting action on the brain. It is showed that blood-letting puncture of Jing-Well points has a certain first-aid effect on the early stage of stroke.

目的 从脑内 c- fos蛋白表达的角度观察井穴放血法对急性缺血性脑损伤的脑保护作用。方法 以凝结法阻断大鼠一侧大脑中动脉造成急性局灶性缺血性脑损伤模型 (MCAO) ,用免疫组织化学法观察缺血区皮层脑组织 c- fos阳性细胞的表达变化及施加手十二井穴放血法后对其的干预治疗作用。结果 急性缺血性脑损伤可快速诱导 c- fos蛋白在皮层缺血区脑组织的表达 ,其表达有随缺血时间延长逐渐增多的趋势 ,而缺血后即刻施加井穴放血的干预治疗 ,在 1h、3h、6 h、2 4 h各缺血时间段均可明显上调缺血区皮层脑组织 c- fos蛋白的表达水平。结论  c- fos蛋白表达既与缺血的严重程度有关 ,也反应神经元的应激反应能力 ,脑缺血能诱导 c- fos蛋白的逐渐表达 ,提示脑的缺血性损伤可应激性地增强神经细胞对缺血、缺氧的耐受和适应能力。而早期施加井穴刺络放血疗法能进一步增强 c- fos蛋白表达 ,提高神经细胞的应激能力 ,从而提高缺血区脑组织的抗损伤修复能力 ,减缓神经细胞凋亡 ,抵抗缺血性脑损害的进一步发展 ,产生有效的脑保护作用 ,表明井穴放血法对中风早期有一定的急救作用。

Objective To observe the brain-protective effects of collateral puncture-bloodletting at JING point on the expression of HSP protein of brain in rats with acute local cerebral ischemia. Methods The cerebral injury with acute local ischemia was induced by occlusion of the middle cerebral artery with blood concretion in rats. The expression of HSP protein in ischemic region of brain tissue was determined with immunohistochemistry and the intervention of bloodletting at JING point was observed. Result Acute cerebral ishemic injury resulted in the expression of HSP protein in the ischemic region of brain tissue and this effect increased gradually with time. Bloodletting at JING point can enhance the expression of HSP protein in initial stage of ischemia and increase the resistance of brain against the exacerbation of the ischemic injury. Conclusion Bloodletting at JING point may improve the anti-stress ability of brain tissue in ischemic field and the protective function of the brain effectively.
