Objective: To investigate the effects of high glucose and heparin on the proliferation and expression of MMP2、MMP9 in normal human mesangial cells (NHMC). Methodology: NHMC was cultured in vitro at the high glucose concentration (HG), with or without heparin, and was controlled by low glucose concentration (LG), with or without heparin. WST-1 was employed to determine the cell proliferation and Western blot analysis was used for the expression of MMP2、MMP9 in NHMC. Results: NHMC proliferation was higher in HG than that in LG medium. They showed a 1.93 fold expansion after 72 hours in HG concentration, in contrast to a 1.63 fold expansion in LG concentration. In the culture with heparin, the proliferation of mesangial cells was inhibited, which was more obvious in HG concentration than that in LG concentration. In HG concentration, with heparin concentration of 50, 100, 200, and 400 mg/ml, the mesangial cells showed a 0.61, 0.52, 0.52 and 0.41 fold reduction in cell numbers compared with that without heparin. In LG concentration, only concentrations of 200mg/ml and 400ug/ml showed reduction in cell number, namely 0.54 and 0.45 fold, compared with that without heparin. In western blot analysis, MMP2 and MMP9 were expressed in both the HG concentration and the LG concentration in NHMC. The expressions increased when heparin was added into HG concentration. Conclusions:①High glucose concentration stimulates the proliferation of NHMC which can be inhibited by heparin. ②Heparin can increase the expression of MMP2 and MMP9 in NHMC, especially at high glucose concentration. ③The renoprotective effects of heparin on diabetic nephropathy may be partially contribute to non-anticoagulation mechanisms.
