AbstractThis study was

作者以对丝裂霉素分别敏感和耐药的人体膀胱癌细胞J82和SCaBER进行对照,研究SCaBER对MMC的耐药机制。药物作用1小时,对SCaBER细胞的IC_(50)是对J_(82)细胞的2.7倍。SCaBER细胞中,两种生物激活酶,即NADPH细胞色素P_(450)还原酶和DT-黄递酶较J_(82)细胞显著降低,提示该细胞对MMC相对低敏感性是由于药物活化过程障碍。BMY25282苯醌还原能力弱,易于活化,其对两种细胞的IC_(50)值相似,进一步支持上述观点。SCaBER细胞的氧自由基形成较J_(82)细胞稍高,抗氧化酶活性无明显变化,提示SCaBER细胞对MMC耐药不是由于氧自由基形成减少。此外,MMC所致SCaBER细胞的DNA链间交联(ISC)较J_(82)细胞明显减少。研究结果表明,SCaBER细胞对MMC耐药是由药物活化障碍和ISC减少所致。

Objective: To study whether the ditribution of lectin receptors in bladder carcinoma are relative to the degree of differentiation and invasion of bladder carcinoma. Methods: Three lectins (Biotin-PNA, Biotin-WGA and Biotin-ConA) were used to study 52 cases of human bladder carcinoma and 10 cases of normal bladder mucosa by using the affinity histochemical method. Results: No PNA and WGA were found in normal bladder mucosa. The positive rate of PNA receptors showed progressively increase in the cases of tumor grade. The statistical analyses showed a significant difference between the above three groups (P < 0 .05 ). The positive rates of PNA and WGA receptors in invasive tumors were significant higher than that in the superficial ones (P < 0.05). Conclusions: The Positive rates of PNA and WGA receptors are ralative to the degrees of diferentiation and invasion of bladder carcinoma.

目的:探讨膀胱癌中凝集素受体分布与其分化程度和浸润深度的关系。方法:应用生物素标记的花生凝集素(PNA)、麦胚凝集素(WGA)及刀豆凝集素(ConA)等3种凝集素对52例人体膀胱癌、10例正常人体膀胱粘膜,进行亲合组织化学法研究。结果:发现正常膀胱粘膜PNA、WGA受体阴性,PNA受体阳性率随膀胱癌病理分级的上升而递增,差异具有显著性意义(P<0.05)。PNA、WGA受体阳性率在浸润性肿瘤中明显高于浅表性肿瘤(P<0.05)。结论:提示PNA、WGA受体阳性率与膀胱癌分化程度和浸润深度有关。
