In this paper we have designed and constructed an antisense gene which is complementary with the assembly origin of TMV-RNA, then a intermediate vector which contains the expressing unitgconsisting of CaMV 35S pro. moter, antisense RNA gene and poly A signal, as well as the NPT-Ⅱ expressing unit was constructed, The NPT-Ⅱ is essential for the selection and transformation of plants, Thus, the successful construction of this interme diate vector will faciliate obtaining the transgenic plants.

With the completion of the sequencing of the yeast genome last year and the sequencing of several other genomes on track, what has been termed ' the post-genomic era looms ever closer, and questions of what this means and where it will lead to become paramount (Editorial, 1997, Nat Genet, 15, 111). Bioinformatics and computational biology are needed for transcrip-tional analysis. Characterization of potential regulatory elements in genomic DNA remains a difficult task. As the analysis of genomes moves into large-scale sequencing, identification and annotation of biologically relevant features in the sequence become increasingly complex and important. A combination of experimental and theoretical approaches will be brought to bear on these challenges.

当前人类基因组研究的重心正在由“结构”向“功能”转移。一个以基因组功能研究为主要内容的所谓“后基因组时代”(post-genomics),即功能基因组学(functional genomics)时代即将到来。功能基因组学主要研究基因的识别及其功能信息的提取、鉴定和开发利用,涉及生物信息学、计算机生物学和生物医学研究等领域。其成果将深化对基因组遗传语言,基因结构与功能的关系,生命的起源与进化,细胞发育、生长、分化,以及疾病发生、发展的分子机理等问题的理解。功能基因组学的研究还将促进生命科学与数学、物理、化学、信息科学、计算机科学以及自动化技术等学科的交叉融汇,刺激相关学科技术领域的发展。

The polymerase chain reaction(PCR) was employed to amplify the full coding region of the BDNF gene from giant panda (Ailuropoda melanoleuca) genomic DNA directly with the primer based on the sequence of human BDNF gene. The amplified fragment was cloned and sequenced.Sequence analysis showed that the coding region of BDNF gene from giant panda was highly homologous with that of human counterpart genes (94% homology) .It was found that there were only two amino acid residues showing difference in the prosequence...

The amplified fragment was cloned and sequenced.Sequence analysis showed that the coding region of BDNF gene from giant panda was highly homologous with that of human counterpart genes (94% homology) .It was found that there were only two amino acid residues showing difference in the prosequence region by comparing the deduced amino acid sequence of BDNF gene of giant panda with that of human. Hower, the amino acid sequence of mature BDNF from giant panda is identical to those of the reported mammalian BDNFs. The succcesful heterologous expression from giant panda of BDNF gene in Escherichia coli was also observed and the molecular weight of the recombinant BDNF precursor was just the same as predicted( ̄28 kD).This is the first report of the functional gene analysis of giant pander at molecular biology level.

参照人BDNF基因序列设计出一对引物和利用聚合酶链式反应(PCR),首次从大熊猫基因组DNA中扩增和克隆到BDNF基因,并使其在大肠杆菌中得到了表达。序列分析表明,大熊猫和人的BDNF基因的核苷酸序列同源性高达94%。在推导的多肽序列中,除在前导肽区有两个氨基酸的差异外,大熊猫BDNF的成熟区与人和其它已报道的哺乳动物BDNF成熟区的氨基酸序列完全一致,显示了极高的保守性。本文首次在分子生物学水平上对大熊猫核基因组功能基因进行研究分析,其结果为深入开展大熊猫的神经系统疾病防治,神经生理以及分子进化,系统发育等方面的研究奠定了重要基础。
