The asymmetric microbiological reduction of the prochiral 13-ethyl-3-methoxy-8,14-seco-△~(1,3,5(10) ,9(11) )-estratetraen-14,17-dione(Ⅲ) with Saccharomyces cerevisiae(2. 346) was sucressful to afford an optical active key intermediate Ⅳ in an excellentyield.Tho D-norgestrel (Ⅰ) and D-13-othyl norgestrienone (Ⅱ) were synthesized fromthis optically active intermediate (Ⅳ) via 8-step and 11-step routes,the total yieldsamounted to 36. 6 and 10%,respectively.

The optically active intermediates 6 (R=CH_3, C_2H_5) for total syntheses of steroidal oral contraceptives was prepared from symmetrical diketones 2(R=CH_3, C_2H_5) by microbial asymmetrical reduction. Thus 2a and 2b were cultivated with Saccharomyces cerevisiae (2.346) to give optically active products 6a and 6b in 73% and 71% yields respectively. The structures and the absolute configurations of these reductive products 6 (R=CH_3, C_2H_5) was established by converting them into the known compounds 4b and 5a...

Thus 2a and 2b were cultivated with Saccharomyces cerevisiae (2.346) to give optically active products 6a and 6b in 73% and 71% yields respectively. The structures and the absolute configurations of these reductive products 6 (R=CH_3, C_2H_5) was established by converting them into the known compounds 4b and 5a respectively, which were obtained from the asymmetrical synthesis of 2 (R=CH_3, C_2H_5) using (-)-(S) proline. A benzene solution of 6 (R=CH_3 or C_2H_5) was stirred for 24 hr with neutral alumina to furnish the optically active compound 15(R=CH_3 or C_2H_5). The formation of the hydroxy compound 15 was postulated as the result of the hydride transfer reaction via a cyclic transition state as shown in fomula "A". The optical purity of 15b determined by using Eu(hfbc)_3 is close to 100%. The R-configuration of C-3' was tentatively assigned by Horeau's method.
