In LHDL-C group,the concentration of HDL-C was correlated negatively with plateleti,β-TG and PF4(all P<0.01). Platelet[Ca2+]i was also correlated with β-TG and PF4 positively in LHDL-C group(all P<0.01).
Methods: We assayed the consentration of platelet [Ca 2+ ]i and the activity of calmodulin CaM respectively of 19 cor pulmonale pateints in their acute stage and of 20 healthy persons, using methods of Fura 2/AM trace labelling with fluorescence and of phosphodiesterase.
These results suggest that the platelet function could be abnormal in VD and ACI patients, and the changes in activity of MLCK and Ca 2+ Mg 2+ ATPase and static density of platelet [Ca 2+ ]i might be involved in the process of molecular pathogenesis of VD.
Conclusion It is suggested that the increase of cytosolic[Ca 2+ i] in platelets following thrombin stimulation should be one of the common mechanism of migraine and cerebral thrombosis the difference of time to attain the maxium level of platelet[Ca 2+ i] levels following thrombin activation between them may reflect the difference of mechanism of two diseases in natural.
Results [Ca2+]i、[Ca2+]ic and CaM levels in patients with acute cerebral thrombosis were (152.25 ± 30.57)nmol/L; (200.36 ± 33.62)nmol/L amd (441.67 ± 209.31)fg/102per platelet respectively, and those in control group were (111.31 ± 21.85)nmol/L;
(158.43 ± 22.44)nmol/L; (301.88 ± 218.10)fg/102 per platelet respectively, [Ca2+]i、[Ca2+]ic and CaM levels in patients with acute cerebral thrombosis were higher than those in the corresponding group of control.
Methods Fura-2 fluorescent labelling technique was applied to measure the [Ca2+]i、[Ca2+]ic and CaM in 3 leases and enzyme linked immunosorbent assay(ELISA) was used to detect the CaM levels in 18 cases of acute cerebral thrombosis.
dauricine significantly inhibited 45Ca2+ uptake into resting and ADP-stimulated platelets in the concentration-dependent way,IC50 were 0.63 and0.29 g·L-1 respectively;
The results showed that resting platelet [Ca2+]i, ADPor thrombin-stimulated platelet elevation amplitude after administration of captopril (12.5 mg, twice daily) for 15 days were significantly reduced in comparison with those before administration.
Using the method of dual-wavelength measurement of platelet [Ca2+]i and Fura-2 as the Ca2+ fluorophore probe, we measured the effect of acidic Mucopolysaccharide fromSticopus Japonicus Selenka (SJAMP) on platelet [Ca2+]i.
The results showed that the most significant increase in platelets [Ca2+]i was seen when the concentration of SJAMP was 100μg/ml and the elevation of normal platelet [Ca2+]i was 93.96 ± 10.24 nmol/L (n=10).
In the presence of extracellular Ca2+ (1 mmol/L), the magnitude of platelet [Ca2+]i response to SJAMP was increased and the [Ca2+]i could reach 116.72±10.
On the other hand, the magnitude of increased platelet [Ca2+]i induced by SJAMP was smaller and the duration of [Ca2+]i reaching the highest level was longer when compared with other platelet aggregation agents.
In this experiment, Disseminated Intravascular Coagulation(DIC) model was Prepared on New Zealand rabbits to observe the change of cytosolic calcium concentration [Ca2 +]i of thrombocytes measured with Quin-2 , in order to study the significance of [Ca2 +]i of thrombocytes in the occurance of endo-toxin-induced DIC. The effect of endotoxin on [Ca2+]i of platelets in vitro was also studied. The results indicated- the resting cytosolic calcium concentration of platelets of New Zealand rabbits is 112±24 nM (n=...
There are several common features between platelets and smooth muscle cells, andthe increment of platelets [Ca~(2+)]i might be a reflection of high arteriolar tension. Following the studyof the relations between nifedipine, indapamide, clonidine and platelets [Ca~(2+)]i, 40 patients (female12, male 28) with essential hypertension devided into 4 groups (EH) were studied with the adminis-tration of capton (captopril, 25mg/d), prazosin (2 mg/d), dihydrochlorothiazide (50mg/d), meto-prolol(50~100mg/d)respective...
Platelet Ca~(2+), erythrocyte Ca~(2+) and Ca~(2+), Mg~(2+)-ATPase activity in erythrocyte membrane of 42 patients of mildmoderate degree heart failure (HF) with diastolic dysfunction were measured. The results showed that the patients with HF had higher level of platelet Ca~(2+) and erythrocyte Ca~(2+) content, and lower level Ca~(2+), Mg~(2+)-ATPase activity than those of control group (P<0.001). There was statistically difference between Ⅰ°andⅡ° HF. Among the heart diseases hypertensive heart disease show...
