This paper reports that Aspergillus vsamii was stimul ated physiologically by dimethyl sulfoxide, resulted in the raising of productivity of citric acid by about 15 percent on labarotary scale, and 6.5 percent on manufacturing scale with the fermentation of solid dregs from sweet potato amylum. The vegetative growth of aerial hyphae was stimulated by dimethyl sulfoxide as its concentration came to about 100-2,000 ppm, inhibited as that came up to 3,000-10,000 ppm, above 4,000 ppm the colour of conidia changed...

Using the glucoamylase-producing strain Aspergillus niger A.S. 3. 4309 as an original strain, a mutagenic strain of Asp.niger 80-72 has been selected by the treatment of UV + DES (diethyl sulferic acid) and then by shaking flask culture.The average glucoamylase activity of five consecutive batches fermentation of strain 80-72 in a five ton fermentation tank was 11327.6 u/ml and the high activity was 12750 u/ml. The average glucoamylase-producing rate was 106.6 u/ml. hr., pH 3.1 at the terminal point of fermentation....

以黑曲糖化酶生产菌A.S 3.4309为出发菌株,用UV+DES复合处理后,经摇瓶筛选,获得诱变株80—72。该菌株在5吨罐连续发酵生产五批,平均酶活力11327.6 u/ml,最高12750u/ml,产酶速率平均106.8 u/ml hr,终点pH3.1;是目前国内黑曲生产糖化酶活力水平最高的优良菌株之一。

The concentration of carbon source and nitrogen source as well as the C/N ratio were significantly affect on the fermentation production of glucoamylase.Results of semicontinuous fermentation showed that the immobilized cells can be used over again for more than 12 times, corresponding to over 30 days. In the first batch fermentation, the fermentation period was 120 hrs and the glucoamylase activity in the broth was almost the same as that produced by free cells. After second batch, the fer-mentation period was shortened from 120 hrs to about 60 hrs and the glucoamylase activity in the broth was higher and higher. The highest glucoamylase activity obtained was 30% higher than that produced by free cells.

黑曲霉As·3.4309的孢子用海藻酸钙凝胶固定化,制成固定化细胞胶粒,用于生产糖化酶的研究。 固定化黑曲霉细胞,产糖化酶的最适温度为32℃,培养基的pH值以5.0为宜。培养基中碳源和氮源的配比对糖化酶的生产有显著的影响。 固定化细胞半连续发酵生产糖化酶的研究结果表明,第一次发酵的周期为120小时左右,发酵液中糖化酶活力接近游离细胞发酵的水平。第二次以后发酵的周期缩短到60小时以下,而产酶水平随着反复使用次数的增加不断提高,最高时达到游离细胞产酶的130%以上。固定化细胞可以连续使用30天。
