Objective: To investigate the relationship between angiogenesis and spontaneous apoptosis of the tumor cell in gallbladder carcinoma. Methods: Forty two surgical specimens from gallbladder carcinoma tissue were studied. Micro vessel density (MVD) was determined by immunohistochemical staining in gallbladder carcinoma tissues with anti human. Factor Ⅷ related antigen (F Ⅷ) antibody. The number of apoptotic cells was measured using in situ end labeling (ISEL) method. Results: The value of MVD was correleted with the depth of invasion (P<0.05), lymph node metastasis (P<0.01) and distant metastasis (P<0.05). ISEL assay demonstrated an inverse correlation between apoptotic index and microvessel densities(r=-0.875,P<0.01). Conclusion: MVD was strongly related to the metastasis of gallbladder carcinoma. The incidence of apoptosis in gallbladder carcinoma was significantly influenced by the extent of neovasculation and angiogenesis may contribute to a reduction of apoptosis in tumor cells.

Apoptosis was detected by eletrionic microscope/flow cytometry/DNA ladder determinations. RT-PCR showed that arsenic trioxide induced up regulation of bax and down regulation of bcl-2.Conclusions In vitro arsenic trioxide can inhibit the growth of QBC939 cell line and induce apoptosis. Its mechanism may correlate with the up-regulation of bax and down regulation of bcl-2.

Objective:To assess the regulatory effect of quercetin on the inflammation,the role of quercetin in the spontaneous apoptosis of neutrophils treated with of lipopolysaccharide (LPS) was investigated.Methods:Apoptosis was determined by light microscopy and flow cytometry.Quantitation and qualitative analysis of DNA fragmentation were performed by diphenylamine assay and agarose gel electrophoresis.Results:Quercetin (1~100 μmol/L) failed to affect the spontaneous apoptosis of neutrophils,but it partly restored LPS delayed neutrophil apoptosis.The concentration of quercetin required to reach a maximum restoring effect was approximately 40 μmol/L.Conclusion:Quercetin prevent LPS induced delay in neutrophil spontaneous apoptosis,which would diminish inflammatory response mediated by LPS primed neutrophils.This could partly explain the anti inflammatory effect of quercetin. 目的 :研究槲皮素对接受细菌脂多糖 (LPS)刺激的中性粒细胞 (PMN)自发性凋亡的影响 ,探讨槲皮素的抗炎作用机制。方法 :以人外周血PMN为研究对象 ,选择光镜和流式细胞术检测细胞凋亡情况 ,对裂解的片段化DNA进行定量 (二苯胺测定法 )和定性 (琼脂糖凝胶电泳 )分析。结果 :槲皮素 (1~ 10 0 μmol/L)对PMN的自发性凋亡率并无影响 ,但却能部分恢复由LPS所延迟的PMN自发性凋亡进程。当槲皮素的浓度为 4 0 μmol/L时 ,其恢复作用达到最大。 结论 :槲皮素对LPS延迟PMN自发性凋亡的效应产生了抑制作用 ,减轻了因预激因子活化PMN而加重的炎症反应 ,部分揭示了槲皮素的抗炎作用机制。
