METHODS: Sprague-Dalwley (SD) rats were randomly divided into three groups: normal control (group N), diabetic nephropathy (group DN), and diabetic nephropathy treated with Irb (group DNI). AIM: To investigate the mechanisms of angiotensin Ⅱ receptor antagonist irbesartan (Irb) in prevention of renal lesion in streptozotocin (STZ)-induced diabetic rats. METHODS: Sprague-Dalwley (SD) rats were randomly divided into three groups: normal control (group N), diabetic nephropathy (group DN), and diabetic nephropathy treated with Irb (group DNI). Diabetes was induced by injection of STZ ip after rats had received uninephroectomy. Blood glucose (BG), body weight (BW), urinary albumin excretion (Ualb), and 24-h proteinuria (24hUpro) were observed in the rats at week 4, 8, and 12, respectively. Creatinine clearance (Ccr), the kidney weight (KW), profile of kidney hypertrophy (KW/BW), renal tissue protein contents (RTP), glomerular area (A_G), glomerular volume (V_G), and width of glomerular basement membrane (GBM) were determined after the rats were sacrificed at week 12. Renal expression of connective tissue growth factor (CTGF) and transforming growth factor-β_1 (TGF-β_1 were determined by immunohistochemistry. RESULTS: There was no significant difference in BG between group DN and DNI (P>>0.05). Irb prevented the increasing of.Ualb excretion, 24 hUpro, and Ccr in diabetic rats (P<0.01). Furthermore, Irb markedly inhibited the increasing of KW, KW/BW, RTP, A_G, and V_G shown in diabetic rats (P<0.05, P<<0.01, respectively). Irb prevented the thickening of GBM and immunostaining of CTGF (P<0.01). The extent of CTGF expression was positively correlated with the glomerular immunostaining for TGF-β_1 and size of V_G (P<0.01). CONCLUSION: Irb exerts an early renal protective role to diabetic nephropathy, possibly through inhibition of renal hypertrophy and expression of CTGF.

Methods Thirty-six Wistar rats were divided into three groups: healthy controls, CCl4 induced cirrhotic rats, and CCl4 induced cirrhotic rats treated with “JinSanE”, which was begun at the fourth week after exposure to CCl4. “JinSanE” was given once a day. Rats were killed after the administration of “JinSanE” for 4 weeks. The anti-fibrosis effect was determined by the expressions of connective tissue growth factor (CTGF) and transforming growth factor-β 1 (TGF-β 1) in liver tissue. The CTGF and TGF-β 1 mRNA expressions were detected by RT-PCR, CTGF and TGF-β 1 were assessed after immunohistochemistry staining. The serum level of hyaluronic acid (HA) was determined by RIA, and the liver histopathology was observed with light and electronic microscopy. Results Compared with the CCl4 induced cirrhotic rats, the expressions of TGF-β 1 and CTGF were decreased in the liver of the rats which were given “JinSanE” (P<0.05 or P<0.01), the serum level of HA was decreased and the histological changes of fibrosis were improved significantly in the rats treated with “JinSanE”. Conclusion “JinSanE” had protective effect against liver injury, and it could inhibit liver fibrosis induced by CCl4 in rats. The effect might possibly be attributed to its anti-inflammatory activity, together with its effect of down-regulating CTGF and TGF-β 1, and suppressing the activation of hepatic stellate cells in rats.

目的 观察金三莪方剂治疗四氯化碳诱导的大鼠肝纤维化的效果及作用机制。方法 将 36只Wistar大鼠随机均分为正常对照组、模型对照组、金三莪治疗组 ,于造模第 4周开始用金三莪方剂治疗 ,第 8周处死全部大鼠。RT PCR检测肝组织的结缔组织生长因子 (CTGF)及转化生长因子β1(TGF β1)mRNA ,免疫组化技术检测CTGF及TGF β1在肝内的表达及其在细胞中的定位 ,放射免疫法检测血清透明质酸 ,肝组织HE染色和电镜检查。结果 与模型对照组大鼠比较 ,金三莪治疗后大鼠肝内CTGF及TGF β1mRNA表达明显降低(P <0 0 1) ;CTGF及TGF β1的免疫反应阳性信号主要位于纤维间隔中的细胞浆 ;治疗后肝纤维化大鼠肝组织内CTGF及TGF β1表达降低 ,肝小叶结构趋于正常 ,纤维间隔变薄。结论 金三莪方剂能有效地减轻四氯化碳诱导的肝纤维化大鼠的肝损伤及纤维化程度 ,其机制可能与抑制肝内CTGF和TGF β1表达有关。

objective To investigate the expression of connective tissue growth factor(CTGF) in rats with unilateral ureteral obstruction(UUO) model and mechanism of selective cyclooxygenase-2 inhibitor celecoxib suppresses tubulointerstitial fibrosis. Methods The UUO models were induced by ligating the left ureter. Forty-five wister rats were randomly divided into celecoxib (celecoxib 100mg/(kg·d) by gastric gavage from 24 h before the obstrction to day 14 after the induction), model and sham operation group. All rats were sacrificed at 3 d ,7 d ,14 d after UUO was initiated respectivety. The expression of transforming growth factor -beta 1 (TGF-β1) ,connective tissue growth factor (CTGF), α-SMA, FN and content of cAMP were examined reverse transcription ploymerase chain reaction (RT-PCR), immunohistochemical stainning and RIA(radioimmuno-assay) respectively. Similar studies were performed in another group of TGF-β1,CTGF were marleedly increased in model group of rats with sham operation. Results The mRNA expression of TGF-β1 and CTGF were markedly increased in another group(UUO 3 d, P< 0.05, UUO 6d and UUO 14d, P< 0.001). With a higher centent of cAMP, the expression of CTGF, α-SMA and FN were inhibited in group celecoxib compared model group, but the expression of TGF-β1 was not different(P >0 05). Conclusions Selective cyclooxygenase-2 inhibitor celecoxib has a salutary effection on tubular and interstitial response to UUO through domn-regulation of CTGF by increasing levels of cAMP.

目的:探讨环氧合酶-2(COX-2)抑制剂塞来昔布(celecoxib)对梗阻性肾病大鼠模型肾组织结缔组织生长因子(CTGF)和转化生长因子β1(TGF-β1)表达的影响及机制。方法:采用单侧输尿管结扎(UUO)建立梗阻性肾病模型,并随机分为模型组和治疗组,另设假手术组作对照。治疗组每日给予塞来昔布100mg/kg,模型组、假手术组每日给予等体积的生理盐水于造模前1d至造模后14d灌胃,分别于UUO后3d、7d、14d分批处死,逆转录聚合酶链式反应(RT-PCR)检测TGF-β1、CTGF的基因表达,免疫组化技术检测TGF-β1、CTGF、α-SMA、FN的蛋白表达,放免法检测组织内cAMP的水平。结果:与假手术组相比,模型组TGF-β1、CTGF的基因表达显著上调,cAMP的水平下降(P<0.001)。塞来昔布对梗阻肾TGF-β1的表达无显著改变,但能升高组织内cAMP水平,抑制CTGF的表达,纤维化程度减轻。结论:在梗阻性肾病大鼠中TGF-β1、CTGF的表达均显著升高,选择性COX-2抑制剂通过升高组织内clip的水平来抑制TGF-B的下游因子———TGF的表达延缓肾间质纤维化。
