To analyze HSP70 expression difference in lymphocytes of soldiers lived in Guangdong(South China),Jiangsu and Ningxia(North China). Flow cytometry was used for detecting HSP70 in peripheral blood lymphocytes of these soldiers after their lymphocytes were incubated at different temperatures: ①basal condition-incubation at 37℃ for 4 h; ②hot damage condition-at 37℃ for 3.5 h,then at 45℃ for 0.5 h; ③cold damage condition-at 37℃ for 3 h,then at 10℃ for 1 h. The expression levels of HSP70 in Ningxia soldiers in the basal and hot damage condition (809.93±58.37 and 959.23±90.28 fluorescence channel,respectively) were significantly higher than those(754.34±60.82 and 887.29±72.85 fluorescence channel)(P<0.05) in Guangdong soldiers. However,the expression levels of HSP70 in lymphocytes of soldiers from the three areas under the cold damage condition did not show significant differences. [Conclusion] The results suggest that there is a significant difference in the expression levels of HSP70 among soldiers from South and North China under the basal and hot damage conditions (P<0.05), but there is no significant difference under the cold damage condition (P>0.05).

[目的 ]分析祖籍在宁夏、江苏、广东三个地区的武警士兵外周血淋巴细胞热、冷损伤时热应激蛋白 70(HSP70 )表达水平的差异性及其可能的意义。 [方法 ]用流式细胞仪检测淋巴细胞中HSP70表达水平。 [结果 ]不同地区比较发现 ,宁夏籍士兵基础和热损伤时 ( 4 5℃ ,0 .5h)HSP70表达水平分别为 ( 80 9.93± 5 8.3 7)、( 95 9.2 3± 90 .2 8)荧光道数 ,均比广东籍士兵分别为 ( 75 4.3 4± 60 .82 )、( 887.2 9± 72 .85 )荧光道数显著升高 (P <0 .0 5 ) ;冷损伤条件下 ( 10℃ ,1h) ,三地区士兵HSP70表达水平差异未见显著性。 [结论 ]南北方武警战士淋巴细胞基础和热损伤条件下HSP70表达水平存在差异性 ;冷损伤条件下 ,三地区士兵HSP70表达水平差异无显著性。

Objective To investigate the changes of plasma adrenaline(Adr) and NOS activity of aortic endothelial cell in cold-exposure rats,so as to provide the basis for medicinal measures to prevent cold injury.Methods L-Arginine(L-Arg) was used to regulate NOS activity of aortic endothelium in cold -exposure rats.Rats were randomly divided into 5 groups: control(room temperatare),cold-exposure Ⅰ(-15 ℃,1 h),cold exposure Ⅱ(-15 ℃,1.5 h),L-Arg(L-Arg 2 g·kg~(-1)),and cold-exposure+L-Arg(-15 ℃,1 h;L-Arg 2 g·kg~(-1)).Blood samples were collectod 6 h after cold exposure.The level of plasma Adr was analyzed by ELISA assay,NOS activity of aortie endothelial cells was measured by nitrate reductase assay,LDH activity in serum and aortic endothelial cell culture media was detected with ultraviolet spectrophotometer, and the level of NOS mRNA in aortic endothelial cells of rats was evaluated by reverse transcription PCR.Results The level of plasma Adr in cold-exposureⅠgroup,and cold-exposureⅡgroup((31.37±4.48),(39.35±5.59) nmol·L~(-1) respectively) was significantly higher than that in control group((10.81±1.85) nmol·L~(-1))(P<0.05).The activity of NOS in endothelial cell of cold-exposure groups((9.19±1.87),(5.94±1.05)μmol·L~(-1) respectively) was significantly lower than that in control group((16.13±3.68)μmol·L~(-1))(P<0.05),while L-Arg could up-regulate it to(12.87±1.60) μmol·L~(-1)(P<0.01),and the level of plasma LDH in cold-exposure group after L-Arg treatment((188.03±36.84) U·L~(-1)) was lower than both cold-exposure groups without L-Arg treatment((244.61±41.63),(365.22±55.19)U·L~(-1))(P<0.01).Conclusion The activity of NOS is inhibited by adrenaline in cold exposure.Up-regulation of NOS activity by NOS promoting agent,L-Arginine,may have some effect to reduce cold injury.
