测定了Langendorff灌流大鼠和兔心脏的31PNMR谱.观测到PCr、ATP、SP、Pi以及PME和GPC等含磷代谢物的共振峰,各谱峰之间实现了很好的分辨.在3~5min的累加时间内测得的图谱具有较好的S/N,可用于以上含磷代谢物的定量测定,以高能含磷化合物ATP和PCr相对峰强度以及冠脉流量的变化对灌流大鼠心脏代谢的稳定性进行了考察,结果表明,在本实验条件下大鼠心脏的能量储存至少可稳定2h,用所建方法以3min的时间间隔对同一大鼠心脏缺血21min及再灌注12min过程中心脏的31PNMR谱进行了连续跟踪测定,并初步观测了缺血及再灌注过程中心肌细胞内ATP、PCr与Pi之间的消长关系.

31P-NMR spectrum of Molt_4 cells consists of resonance peaks of phosphomonoester,inorganic phosphate (Pi),diphosphodiesters and ATP. Changes in phosphorus containing compound after action of interferon_γ on Molt_4 cells were observed by 31P-NMR spectrum. The pH in intra_molt_4 cells were monitored based on chemical shift of Pi. Several intracellular phosphorus containing compounds can be simultaneously monitored by 31P-NMR.

Molt_4细胞的31P核磁共振谱由磷酸单酯、无机磷(Pi)、双磷酸双酯、三磷酸腺苷(ATP)的共振峰组成。用31P核磁共振观察了γ-干扰素对Molt_4细胞作用后含磷化合物的变化。根据Pi的化学位移对Molt_4细胞内pH进行了监测。31P核磁共振能一次同时监测细胞内多种含磷代谢物,是一种无损伤研究细胞代谢的有效方法。

Ischemic preconditioning (IPC) will protect the heart from the damage caused by a subsequent long ischemia period. 31 P spectra of isolated perfused rat heart measured by the nuclear magnetic resonance (NMR) surface coil technique can be used to continually, dynamically and noninvasively obtain metabolism information. The ATP and PCr recovery rate of the IPC group after ischemia is significantly higher than that of the control group. In conclusion, the above results support that IPC can protect the rat heart by reducing damage during the ischemia period.

缺血预处理 (IPC)是一种肌体对长时间缺血产生耐受性的处理技术。应用核磁共振表面线圈技术获得大鼠心肌在灌流的不同阶段的 31 P谱 ,可以从能量代谢的角度对缺血 /再灌注心脏进行连续、动态、无损伤的检测 ,从含磷代谢物的转化对缺血预处理产生作用的机理进行研究。实验结果显示 ,缺血预处理的保护作用并不是通过有利于心肌内 ATP和 PCr(磷酸肌酸 )恢复的水平来体现的 ,而是在缺血期通过缓和 p Hi(胞内 p H)值的突然变化和 ATP的大量消耗以及通过 p Hi 值和 ATP的尽快恢复来减轻心肌组织结构在缺血期产生的严重损伤 ,从而有利于再灌注时心肌功能的恢复
