Objective To investigate the micro anatomy of the sciatic nerve of adult rats and to probe its significance in the established model of denervated gastrocnemius muscle. Methods Micro dissection and observation were done in 120 sciatic nerves of 60 rats and sciatic nerves of another 48 rats were sectioned at different levels to observe the occurrence of ulcers in the affected limbs and measured the wet weight of the corresponding gastrocnemius and the diameter of the muscle cells. Results 1. The sciatic nerve was composed of the fourth, fifth and sixth lumbar and the first sacral nerve roots. The blood circulation was supplied by inferior gluteal artery, internal and external pudendal artery, perforating branches of femoral artery and popliteal artery. The mean transverse and anterior posterior diameters of sciatic nerve at the levels of ischial tuberosity, middle one third and lower one third of the thigh were 1.54mm, 1.53mm, 1.52mm and 0.91mm, 0.89mm, 0.88mm respoectively. The retrograde dissection length of tibial nerve was 1.5cm and forced dissection length was 2.0cm. The gastrocnemius muscle was innervated by the medial and lateral muscular branches of the posterior tibial nerve. 2. The limb ulceration rates were 33.3%, 25% and 8.3% in the groups of sciatic nerve sectioned at the levels of ischial tuberosity, lower one third of the thigh and tibial nerve section respectively. There was no ulceration in the group of which the nerve to the gastrocnemius was sectioned. There were no signifiicant differences of the muscle wet weight and the muscle cell diameter in all three groups. Conclusion The sciatic nerve of adult rat can well be used for experimental peripheral nerve injury and repair model, while in establishing a model of denervated skeletal muscle, either sectioning of tibial nerve or its branch to the muscle would be more appropriate. 目的研究大鼠坐骨神经显微解剖结构,探讨其对建立大鼠失神经支配腓肠肌模型的意义。方法先对60只大鼠120条坐骨神经进行完整显微解剖学观察;再将48只大鼠坐骨神经在不同平面切断,建立失神经支配腓肠肌实验模型,观察肢体溃疡情况,测量腓肠肌湿重及肌细胞直径。结果1.大鼠坐骨神经主要由腰4,5,6及骶1神经根组成;血供来自臀下动脉,阴内、外侧动脉,股动脉穿支及动脉分支;在坐骨结节、大腿中1/3及下1/3平面的横径及前后径分别为1.54mm、1.53mm、1.52mm及0.91mm、0.89mm、0.88mm;胫神经逆行可分离长度及强行分离长度分别为1.5cm及2.0cm;腓肠肌由胫后神经内、外侧肌支支配。2.在坐骨结节平面、大腿下1/3切断坐骨神经主干及切断胫神经,肢体溃疡发生率分别为33.3%、25%及8.3%,切断腓肠肌支无溃疡发生;各组腓肠肌湿重及肌细胞直径无显著性差异,P>0.05。结论大鼠坐骨神经是建立周围神经损伤与修复的良好实验模型,在建立失神经支配骨骼肌模型时,以切断腓肠肌支或胫神经为宜

Objective To study the histology,electro-physiology and enzymic histochemistry of denervated skeletal muscle to find out the index evaluating the degree of muscle atrophy.Methods Forty-eight SD rats were used and the model of denervated skeletal muscle was made by cutting off the right tibial nerve.The myocyte diameter and cross section area,motor end plate,fibrillation potential amplitude,Na +,K +-ATPase and Ca 2+ -ATPase activities were observed.Results The myocyte diameter and cross section were reduced progressively along with the duration of muscle denervation.The change of motor end plate was not significant within 4 weeks and disappeared after 16 weeks.The fibrillation potential amplitude could remain at a higher level within 8 weeks and reduced progressively afterwards.Na +,K +-ATPase activity was reduced progressively along with the duration of denervation.Ca 2+ -ATPase activity could maintain at a higher level within 8 weeks and was reduced significantly afterwards.Conclusion The myocyte diameter and cross section area was a credible morphologic index of muscle atrophy.The fibrillation potential amplitude could be considered as a potential electric physiologic index.Na +,K +-ATPase-ATPase activity was a credible enzymic histochemisty index.

目的 观察大鼠失神经支配骨骼肌组织学、电生理及酶组织化学改变 ,探讨反映肌肉萎缩程度的指标。方法 选用SD大鼠 ,建立失神经支配腓肠肌模型 ,观察术后肌细胞直径及截面积 ,运动终板超微结构、纤颤电位波幅、Na+ 、K+ ATP酶及Ca2 + ATP酶活性变化。结果 肌细胞直径及截面积随失神经支配时间延长呈进行性下降 ;运动终板 4周内改变不明显 ,16周后消失 ;纤颤电位波幅在 8周内维持在高水平 ,12周后呈进行性下降 ;Na+ 、K+ ATP酶活性随失神经支配时间延长呈进行性下降 ;Ca2 + ATP酶活性在 8周内维持在较高水平 ,12周后明显下降。结论 肌细胞直径及截面积可作为反映肌肉萎缩的可靠形态学指标 ,纤颤电位波幅可作为潜在电生理指标 ;Na+ 、K+ ATP酶活性是可靠酶组织化学指标。

Objective:To establish two-dimensional gel electrophoresis(2-DE) system for skeletal muscle, and find differences of gastrocnemius protein of denervated rat and control group.Methods:Establish the model of denervated gastrocnemius, extract the total proteins from the denervated gastrocnemius tissue and normal gastrocnemius tissue, and run isoelectric focusing(IEF) electrophores as the first dimension , the sodium dodecyl sulfate polyacrylamide gel electropheresis(SDS-PAGE) as the second dimension to separate the total proteins . Protein spots were visualized by Bio-safe Coomassie and the image were abtained through the GS-800 scanning. The protein spots were analyzed by PDQuest software and measure the position deviation of matched spots in IEF and SDS-PAGE.Results:For normal gastrocnemius tissue, the average protein spots of 3 gels were 560±16; and 471±19 spots were matched with the average matching rate of 84.1%. For denervated gastrocnemius tissue, the average protein spots of 3 gels were 545±13; and 447±17 spots were matched with the average matching rate of 82.1%.A total of 445±8 spots were matched between the electrophoretic maps of 3 denervated gastrocnemius tissue and paired normal gastrocnemius tissue. A good reproducibility of spot position 2-DE map was obtained. 80 spots were found significantly changed in quality and quantity in the group of denervation. The average position deviation of matched spots was 1.203±0.763 mm in IEF direction, and 1.062±0.529mm in SDS-PAGE direction.Conclusions:There are distinctive differences between 2-DE maps of denervated gastrocnemius and control group,downregulated proteins may be involved in keeping the normal function of muscle and upregulated proteins may participate in the process of atrophy after denervation.

目的:建立骨骼肌蛋白分离的双向电泳(2-DE)技术体系,寻找失神经支配骨骼肌与正常骨骼肌蛋白的表达差异。方法:建立失神经腓肠肌模型,提取腓肠肌总蛋白,进行第一向等电聚焦(IEF)和第二向十二烷基硫酸钠聚丙烯酰胺凝胶电泳(SDS-PAGE)技术分离总蛋白,使用PDQuest软件对凝胶图像进行分析,并测量了凝胶蛋白斑点在IEF和SDS-PAGE方向上的位置偏差。结果:失神经组和正常组腓肠肌组织3块凝胶的平均蛋白质点数分别为560±16和545±13,平均匹配点数分别为471±19和447±17,匹配率达84.1%和82.1%。对比分析了两种腓肠肌组织的双向电泳图谱发现,平均匹配点数为445±8;发现在失神经支配后有80个点发生了明显而稳定的质和量(大于10倍或小于0.1倍)的改变。不同凝胶间蛋白质点在IEF方向的偏差为(1.203±0.763)mm,在SDS-PAGE方向上的偏差为(1.062±0.529)mm。结论:失神经组和正常组腓肠肌的双向电泳图谱存在明显差异,失神经后表达下调的蛋白可能与维持肌肉正常功能相关,而表达上调的蛋白则可能与肌萎缩的发生过程相关。
