Synthesis and Evaluation of Homologous Analogs of Anticonvulsant 5-Substituted 3,3-Diethyl-4,5-Dighdro-2(3H)-Furanones
      
Novel 5-substituted esters and homologous ester and amido derivatives of 4,5 dihydro-3,3-diethyl-2(3H)-furanone were synthesized and evaluated for anticonvulsant activity in rodents and for affinity to a site on the GABAA receptor complex ([3H]TBOB).
      
We studied the interaction between O,O-diethyl-S-[(N-acyl-N-alkoxycarbonylalkyl)aminomethyl]thiophosphates and mammalian cholinesterases as well as esterases from insect tissue extracts by kinetic methods and disc electrophoresis.
      
Using selective chemical modification by diethyl pyrocarbonate (DEP), the possible involvement of His residues in this interaction was studied.

The reactions between O,O-diethyl-dithiophosphoric acid and diethylmercury,and between

二硫代磷酸 O,O-二烃基酯或其汞盐与二乙基汞很顺利地进行下列反应:(?) (?)这两个反应可利用来制备各种二硫代磷酸 O,O-二烃基酯乙基汞盐。这里制备了九种同系物。在报告Ⅱ中所制备的二硫代磷酸 O,O-二β-萘基酯,表现有2,4-D 类型的植物刺激作用,双-(O,O-二烃基硫磷酸酯)二硫物的杀虫(粘虫三龄幼虫)及杀菌(Alternariatenius)效能均甚微弱。但是本报告中所制备的二硫代磷酸 O,O-二烃基酯乙基汞盐同时具有杀虫(粘虫的三龄幼虫)及杀菌(Alternaria tenius)的效能。其中有六种,如以含汞量计,它们的杀菌效能较氯化乙基汞为强。

(1) Eight dialkylphosphites (I), including, dimethyl-, diethyl-, di-n-propyl-,di-iso-propyl-, di-n-butyl-, di-iso-butyl-, di-sec-butyl-, and di-n-amyl- phosphite,reacted with aniline in the presence of carbon tetrachloride and a tertiary baseto form the corresponding dialkylanilinophosphonates (II). It was found thatthe reaction became increasingly difficult for the dialkylphosphites in thefollowing order: dimethyl-, diethyl-, di-n-propyl-, di-n-butyl-, di-n-amyl-,di-iso-butyl-, di-iso-propyl-, and di-sec-butylphosphite....

(1) Eight dialkylphosphites (I), including, dimethyl-, diethyl-, di-n-propyl-,di-iso-propyl-, di-n-butyl-, di-iso-butyl-, di-sec-butyl-, and di-n-amyl- phosphite,reacted with aniline in the presence of carbon tetrachloride and a tertiary baseto form the corresponding dialkylanilinophosphonates (II). It was found thatthe reaction became increasingly difficult for the dialkylphosphites in thefollowing order: dimethyl-, diethyl-, di-n-propyl-, di-n-butyl-, di-n-amyl-,di-iso-butyl-, di-iso-propyl-, and di-sec-butylphosphite. Seven auxiliary bases, namely, triethanolamine, methylpiperidine, 2, 4, 6-col-lidine, α-picoline, pyridine, dimethylaniline and diethylaniline, were employedin above reaction. The auxiliary effects of the tertiary bases used decreasedin the following order: triethanolamine, methylpiperidine and 2, 4, 6-Collidine;while α-picoline, pyridine, dimethylaniline and diethylaniline gave no auxiliaryeffect. Obviously the basicity of auxiliary base was very important. (2) This method can also be used to prepare dialkyl phenylhydrazino-phosphonates (IV), such as diallyl-, dimethyl-, diethyl-, di-iso-propyl-, di-iso-butyl-, di-n-amyl-, di-iso-amyl-, ethyl n-butyl- and ethyl n-amyl-phenylhydrazinophosphonates.

Alkaline and acid hydrolyses of polyacrylamide have been studied.Hydrolyses caused by dilute alkali or dilute acid were found to beincomplete, while those by concentrated alkali and acid approaching completeness.In the range of 0.6--3.3 N sodium hydroxide and at 100℃, the limiting degreeof hydrolysis is around 65--70%. It was found that Van Slyke method couldbe applied to the quantitative determination of the amide group in the polymerswhile the nitrile groups causing no interference. Alkaline and acid hydrolyses of polyacrylamide have been studied.Hydrolyses caused by dilute alkali or dilute acid were found to beincomplete, while those by concentrated alkali and acid approaching completeness.In the range of 0.6--3.3 N sodium hydroxide and at 100℃, the limiting degreeof hydrolysis is around 65--70%. It was found that Van Slyke method couldbe applied to the quantitative determination of the amide group in the polymerswhile the nitrile groups causing no interference. The identity period ofpolyacrylamide was determined to be 5.2A through X-ray analysis. Copolymers of acrylamide with acrylic acid, allyl alcohol or vinyl acetatehave been synthesized and their behavior towards alkaline hydrolysis has beenstudied. The data for the completeness of the hydrolysis of the latter twocopolymers served to support the suggestion that the screening or repulsiveeffect of the neighboring negatively charged carboxylate groups played animportant role in hindering the hydrolysis of polyacrylamide from going tocompleteness. Several N-alkylsubstituted polyacrylamide and their copolymers have beensynthesized. Their degrees of hydrolysis were determined in the followingorder of completeness: polyacrylamide> poly N-methyl acrylamide> polyN, N-dimethylacrylamide> poly N, N-diethylacrylamide. It was pointed outthat besides the above stated screening effect of negatively charged groups, sterichindrance of N-alkyl- and N, N-dialkylamino groups in the polymer moleculesshould never be neglected.

本文报告聚丙烯酰胺的硷性水解及酸性水解。用浓酸或浓硷处理,都可接近完全水解,用稀酸或稀硷水解则不完全。在0.6~3.3N氢氧化钠及100°时,水解程度为65—70%,Van Slyke法可用以测定丙烯腈与丙烯酰胺共聚物中酰胺基量。用X-射线衍射法测定聚丙烯酰胺长链的等同周期为5.2A。硷性水解之不完全,主要是由于邻近羧基的负电场对羟基向酰胺基进攻时的屏蔽作用所致。合成丙烯酰胺分别与丙烯酸、丙烯醇、或乙酸乙烯酯的共聚物,从而研究其硷性水解,前者的水解限度为75%,后二者可以接近完全,直接证明邻近非负性基团不能产生负电场因而没有屏蔽作用,故水解接近完全。研究若干N取代聚丙烯酰胺及共聚物的水解,在一定条件下它们的水解程度按下列次序递减:聚丙烯酰胺>聚N-甲基丙烯酰胺>聚N,N-二甲基丙烯酰胺>聚N,N-二乙基丙烯酰胺。除了邻近负电场的屏蔽作用外,N及N,N-取代基在长链中有不容忽视的空间阻碍。
