Objective:To study the effect of Zhu Mu Gen San(a kind of Tibetan compound recipe)on the memory disorders of rat's with cerebral ischemia and its affect the content of NE in rat′s hippocampus.Methods:The cerebral ischemia model was made by the occlusion of the rat's bilateral carotid arteries.To detect the rat's memory ability,the rat's behavior was tested by the water maze and the electric maze before and after feeding drugs.The high performance liquid chromatography with fluorometric detection was carried out to observe the norepinephrine (NE) level changes in rat's hippocampus after feeding drugs.Results:Compared with the control group,the rat's hippocampus NE level increased markedly (P<0 05)in Zhu Mu Gen San treated group;the time which the rats passed through the maze reduced parkedly (P<0 05).It indicated that the rat's memory ability had improved remarkably. Conclusion:Zhu Mu Gen San could improve the memory disorders in rat's with cerebral ischemia.The mechanism might relate to its effect on the content of NE in rat's hippocampus.

目的 :研究珠穆根散 (一种藏药复方 )对急性脑缺血大鼠所致记忆障碍与海马去甲肾上腺素 (NE)的含量之间的关系。方法 :通过夹闭大鼠两侧颈总动脉 ,造成了大鼠急性脑缺血模型。采用水迷宫和电迷宫 ,在喂药前和喂药结束后对大鼠进行行为检测以检验大鼠记忆能力的变化。喂药结束后 ,用高效液相色谱———荧光检测法测定了各组大鼠海马组织去甲肾上腺素 (NE)的含量变化。结果 :喂珠穆根散 5 0天后 ,药物治疗组与治疗对照组相比 ,大鼠通过迷宫的时间显著缩短 (P <0 0 5 ) ,说明其记忆力有明显的改善 ;大鼠海马去甲肾上腺素含量明显增高 (P <0 0 5 )。结论 :藏药珠穆根散对急性脑缺血所致大鼠记忆障碍有明显的改善作用 ,可能是与其影响海马内去甲肾上腺素的含量有关

目的探讨补肾填精方防治铝致阿尔茨海默病(AD)的分子机制。方法用长期腹腔注射AlCl3复制大鼠动物模型,用RT-PCR检测端脑皮质beta-淀粉样前体蛋白mRNA异构体-APP695mRNA水平,用Y电迷宫仪检测大鼠的学习记忆能力。结果模型组大鼠端脑皮质APP695mRNA的表达水平明显升高(P<0.05),学习记忆能力明显降低(P<0.05)。中药干预后,端脑皮质APP695mRNA表达较模型组明显降低(P<0.01),学习记忆能力明显提高(P<0.05)。结论端脑皮质APP695mRNA表达水平升高与学习记忆能力减低明显相关,通过抑制端脑皮质APP695mRNA的过量表达是补肾填精方防治AD模型大鼠学习记忆能力损害的机制之一。

AIM: To observe the improvement function of huperzine A transdermal drug delivery system (Hup A TDDS) administration at Governor Vessel(GV) acu points on memory disorders induced by scopolamine in mice and to compare the effect of GV acu points administration with general place administration. METHODS: Step down test and "Y" maze test were used to observe the influence of Hup A TDDS administration at GV acu points on memory disorders induced by scopolamine. Some biochemical indices were also determined....

AIM: To observe the improvement function of huperzine A transdermal drug delivery system (Hup A TDDS) administration at Governor Vessel(GV) acu points on memory disorders induced by scopolamine in mice and to compare the effect of GV acu points administration with general place administration. METHODS: Step down test and "Y" maze test were used to observe the influence of Hup A TDDS administration at GV acu points on memory disorders induced by scopolamine. Some biochemical indices were also determined. RESULTS: (1) The marked effects were found among 3- 6 h after Hup A TDDS administration at GV baihui acu point. The best effects showed 4 h after administration, and it weakened a little from 5 h to 6 h in 3.4 μg/ 0.25 cm 2 group. At 26.7 μg/ 0.25 cm 2 group, the drug action maintained an effective level from 3 h to 6 h, and reached the maximum at 4 h. (2) Among 5 acu points of GV to administration at Hup A TDDS 26.7 μg/ 0.25 cm 2, the obvious effects on memory disorders existed, and baihui (GV20) was the most effective acu point. (3) The administration of Hup A TDDS ( 26.7 μg/ 0.25 cm 2 and 13.4 μg/ 0.25 cm 2) through both acu point and general place (buttocks), approved memory improvement function by step down test and "Y" maze test, and the action in baihui acu point group showed more significant. The depressed trend of cholinesterase activity in brain was proved in all of both GV acu point and general place groups and baihui acr point group exposed signal difference when compared with mokcl group. CONCLUSION: Hup A TDDS administration at GV acu point shows a better improvement of memory disorders in comparison with general place. The action mechanism may be due to the depressed cholinesterase activity and the improved cholinergic system function.
