Objective To investigate the mechanism of Chinese medicine for treating acne and the pharmacological effect of Cuochuang Granules(CG).Methods The test of bacteriostatic in vitro,antibacterial test in vivo were carried out,and the effects of CG on acute exudative inflammation of mouse auricle,on mouse abdominal capillary permeability,on rat abdominal leukocytoplania,on microcurculation of mouse auricle and on uterus weight in female deovary mice were observed in Kunming mice and Wister rats.Results CG had the bacteriostatic effect in different degrees in vitro on 47 strains of 5 species of peccant bacteria,especially on Staphylococcus aureus,Staphylococcus epidermidis and acne diphtheroid.CG had a protective effect in vivo on mouse abdominal inflammation of Staphylococcus aureus.The survival rate of mouse was 70%.CG had an obvious inhibitory effect on acute exudative inflammation of mouse auricle induced by dimethylbenzene(P<0.01).CG had an inhibitory effect on accentuation of capillary permeability of mouse abdomen induced by acetic acid.The inhibitory rate of WBC number was 62.65% in the inflammatory transudate induced by CMC.CG could dilate capissary arteries and veins of mouse auricle at different times.The comparison to the blank group was P<0.01 or P<0.05.CG increased the uterus weight of female deovary mouse(P<0.05). Conclusion CG has the obvious effects of bacteriostatic,antiinfection and microcirculation improvement.These effects are consistent with CG's effects of cooling blood to remove toxin,drying dampness and clearing away heat,and activating blood to subside swelling.

目的探讨中药治疗痤疮机理,了解痤疮颗粒的主要药理作用。方法选择昆明种小鼠和W istar大鼠,分别进行体外抑菌、体内抗菌、对小鼠耳壳急性渗出性炎症的影响,对小鼠腹腔毛细血管通透性的影响,对大鼠腹腔白细胞游走的影响,对小鼠耳廓微循环的影响和对去卵巢雌性小鼠子宫重量的影响实验。结果痤疮颗粒对5种47株临床致病菌有不同程度的体外抑菌作用,其中对金黄色葡萄球菌、表皮葡萄球菌、丙酸痤疮杆菌的生长有较强的抑制力;痤疮颗粒对腹腔感染金黄色葡萄球菌的小鼠有明显的保护作用,存活率为70%;对二甲苯致小鼠耳壳急性渗出性炎症,有明显的抑制作用,P<0.01;对醋酸所致小鼠腹腔毛细血管通透性亢进有一定抑制作用;对羟甲基纤维素(CMC)引起的炎症渗出液中白细胞数量抑制率为62.65%;不同时间扩张小鼠耳廓细动脉和细静脉,与空白对照组比较P<0.01或P<0.05;对去卵巢雌性小鼠子宫重量明显提高,P<0.05。结论痤疮颗粒具有明显抑菌、抗炎、改善微循环的作用,该作用与本药凉血解毒、燥湿清热、活血消肿功效相一致。

To develop an RP-HPLC method for the determination of baicalin content in furuncie granules.Methods:The hypersil C_(18) columm and the mobile phase of methanol-water-phosphoric acid(41∶59∶(0.25)) were used.The flow rate 1.0ml/min and the detective wave length 276nm.Results:The average recovery was 99.38% and the related standard deviation(RSD)was 2.48%(n=5).Conclusion:The method was quick,accurate,and could be used to control its quality.

目的:应用反相高效液相色谱法测定痤疮颗粒剂中黄芩苷含量。方法:采用C18柱,流动相为甲醇-水-磷酸(41∶59∶0.25),流速为1.0ml.min-1,检测波长276nm。结果:平均回收率为99.38%,相对偏差RSD为2.48%(n=5)。结论:方法快速,精确,可作为该药质量控制方法。

目的:研究痤疮颗粒的作用机理。方法:用放射免疫法测定大鼠血清睾酮浓度,测算大鼠睾丸、附睾、精囊、前列腺系数,观察痤疮颗粒对性激素的影响;分别进行二甲苯致小鼠耳廓肿胀实验,醋酸所致小鼠毛细血管通透增加实验,观察该药的抗炎作用。结果:痤疮颗粒能够降低丙酸睾酮(TP)引起的未成熟大鼠血清睾酮的升高,降低其附睾、精囊、前列腺的重量;降低醋酸所致小鼠腹腔毛细血管的通透性;减轻二甲苯所致小鼠耳廓的肿胀。结论:痤疮颗粒能够调节体内激素分泌,缓解二甲苯致小鼠耳廓肿胀程度,减轻醋酸所致小鼠毛细血管通透性增加现象。
