ObjectiveTo investigate whether retinal ganglion cell death in experimental glaucoma is induced by apoptosis.MethodsChronic elevated eye pressure was produced by injecting fixed erythrocytes into the anterior chambers and the rabbits were dilled at 7,14,21 and 28 d postoperatively.Analytic methods included electron microscopy and immuno histochemistry staining for DNA fragmentation(TUNEL method).ResultsDead ganglion cells in experimental retinas exhibited morphologic features of chromatin condensation.Cells with a positive reaction for DNA fragmentation were observed in eyes subjected to experimental glaucoma and none in control eyes.ConclusionThe death of retinal ganglion cells in rabbits with experimental glaucoma was through apoptosis.It may be possille to treatment glaucomatous damage to the retina by the potential interrupution of the program cell death.

目的 研究兔的实验性青光眼视网膜神经节细胞的死亡是否有凋亡参与。方法 前房注入固定的红细胞悬液使兔眼压升高 ,分别在术后 7,14 ,2 1,2 8天处死动物 ,取视网膜组织 ,TUNEL标记染色 ,电镜观察。结果 电镜下可见视网膜神经节细胞死亡特征为典型的凋亡早期特征—核浓染。免疫组化TUNEL法实验组发现神经节细胞凋亡 ,而正常对照组没有发现。结论 实验性青光眼的视网膜神经节细胞的死亡有凋亡参与。这为通过调控凋亡而治疗青光眼的视网膜视神经损伤提供可能

Objective To detect the expression on p53 gene in retinal ganglion cells(RGCs) of rabbit eyes with different degrees of intraocular hypertension. Results The number of RGCs with a positive reaction for P53 monoclonal antibody was statistically higher in the experimental eyes than in their control fellow eyes(P < 0. 01 ),and apoptosis was mainly found in the early stage of research. Conclusion Our research presents a new alternative view of apoptosis in experimental glaucoma(According to former reports,apoptosis was mainly found in the advanced stage of the patients with POAG).

目的检测不同眼压状态下实验性青光眼视网膜神经节细胞(RGCs)p53基因的表达。方法家兔16只,任选 1眼作实验,另眼对照。 2%甲基纤维素0. 10~0. 15 ml前房注射制做高眼压模型,根据眼压值低、中、高不同对动物进行分组。分期取材、石蜡切片、免疫组化染色检测 p53蛋白的表达情况。采用计算机图像分析技术对 RGCs密度进行定量分析。结果凋亡率高低依次为:眼压较低组(1.86%)>眼压中等升高组(0.93%)>眼压较高组(0.53%)>对照组(0.05%)(P<0.01)。结论与前人对POAG患者的研究结果相反,在眼压急速升高的模型眼中,凋亡主要发生在实验的早期。
