By using of NADPH-diaphorase histochemical method,the localization of nitric oxide synthase(NOS)in the major pelvic ganglion(MPG)and penile erectile tissue of the rat was investigated.A large percentage of the neurons in MPG was NOS positive,most of the neurons were concentrated on one end of the MPG,where the pelvic nerve entered.The penile deep arteries and their branches,helicine arteries in penile corpus were surrounded by NOS-positive fibers.NOS-positive fibers were seen also in the bundles of smooth muscles in the wall of the cavernous sinus of both penile and urethral corpus.No NOS-positive cell bodies appeared in penile erectile tissue,After fluoro-gold(FG) was injected into penile erectile tissue,many FG-labeled neurons were found in the MPG,Further FG tracing method combined with NADPH-diaphorase histochemistry indicated that almost all of FG-labeled neurons in the MPG were NOS-positive, It is concluded that NOS-positive neurons were distributed in the MPG,and a part of which innervated vascular beds of penile erectile tissue.The present results suggested that nitric oxide rnay be an important neuroactive substance for erection of penis.

目的研究降钙素基因相关肽(CGRP)和硝普钠(SNP)对阴茎组织和血管的舒张效应及其抑制反应。方法离体组织灌流法。结果CGRP对犬阴茎勃起组织具有不同程度的舒张效应,其作用可为左旋硝基精氨酸(L-NNA)和亚甲蓝(MB)大部分抑制;SNP具有更强烈的舒张效应,但仅为MB抑制,不为L-NNA抑制。结论CGRP舒张阴茎海绵体平滑肌和阴茎血管的作用可能部分地通过左族精氨酸-一氧化氮-环磷酸乌苷通路而实现。

Objective To investigate the effect of cavernous nerve injury on nNOS containing nerve fibers in corpus cavernosum. Methods The quantity of nNOS positive nerve fibers in rat erectile tissue were examined with immunohistochemistry techniques (SP method )after cavernous nerve injury. Results After bilateral ablation,the nNOS positive nerve fibers were significantly decreased at 3 weeks (17±4) and remained so at 6 months (16±4).For unilateral injury group,the nNOS positive nerve fibers were similarly decreased on the side of neurotomy at 3 weeks (18±6),but by 6 months,the number increased significantly (61±9) and approximated the level on the contralateral side (81±13). Conclusions It was demonstrated the regeneration of nNOS containing nerve fibers may occur 6 months after unilateral cavernous nerve ablation,while this process didn't happen after bilateral cavernous nerve injury.It suggests that cavernous nerve has to be preserved at least one side during pelvic radical surgery.

目的 探讨海绵体神经损伤对阴茎海绵体nNOS神经纤维的影响。 方法 采用免疫组织化学技术SP法测定大鼠海绵体神经损伤后阴茎勃起组织nNOS神经纤维数量。 结果 双侧损伤组术后 3周 ,阴茎海绵体nNOS神经纤维显著减少 ,6个月后仍很少 ;单侧损伤组 ,术后 3周损伤侧nNOS神经纤维的改变类似于双侧损伤组 ,但 6个月后损伤侧nNOS神经纤维明显增多 ,接近于对侧水平。 结论 阴茎海绵体nNOS神经纤维在单侧海绵体神经损伤后可能会再生 ,而双侧海绵体神经损伤后则不能再生 ,提示在进行盆腔根治性手术时 ,要保留一侧海绵体神经 ,以维持勃起功能
