Two samples of definite volume of supernatant are taken, and after addition of standard acid or base to make the solution 0.10N with respect to either acid or base, optical densities are measured at 249, 262.5, 271, and 290mμ. for acid solutions, and 258, 262, 282.5, and 290mμ, for basic solutions. The amount of each of the purine and pyrimidine bases is then calculated by the following equations:T=0.231·△D_((290)_(13)-(290)_1)·VG=0.192·△D_((249)_1-(258)_(13)·V-0.325TC=0.150·△D_((271)_1-(262)_(13)·V-0.466TA=0.105·△D_((262.5)_1-(282.5)_(13))·V-0.317TA, G, C, and T represent the micromole of the bases; V, being the total volume (in ml) of the solution.
