Amylase,lipase and trypsin activities of the pancreas in the different stages of the secretory cycle in mice were determined by Somogyi's,Archibold's and Willsttter's methods.The corresponding changes of the zymogen granules were shown by Lacy's modification of Baker's Sudan black method.Histochemical observations on lipase,RNA and DNA were also made. The experimental animals were divided into three groups: (1) those after 24 hours starvation;(2) those after receiving 4 injections intraperitoneally of pilocarpine 0.014 mg/g tissue at 30 minutes intervals;(3) those after recovering for a certain period(6,12,18 or 24 hours)following the pilocarpine injection. The results show that there is a definite correlation between the number of zymogen granules in the acini and the content of amylase,lipase,and trypsin in the pancreatic tissue.The highest content of these three enzymes was found in the resting stage of the pancreas.The activity of these three enzymes decreased to a minimal level after the in jections of pilocarpine,requiring about 12 hours of recovery to return to the resting value. The zymogen granules are shown to accumulate in the cells until they are absorbed by the secretion bodies.The recovery phase appears to start with the onset of the zymogen discharge,since both prozymogen bodies and secretion bodies have been obsefved together in the same cells.The recovery phase,however,is maximal when all the secretion bodies have been extruded,for then the cells contain many prozymogen bodies Eventually the number of prozymogen bodies decreases as the number of zymogen granules increases.The cells then gradually enter into the resting phase and contain few prozymogen bodies and many zymogen granules.It is seen that during the secretory cycle a striking parallelism exists becween the morphological changes of the zymogen granules and the changes of the activity of the three enzymes.The RNA and DNA content of the pancreas shows no change during the cycle. The changes of the lipase activicy demonstrated by the histochemical method in the different stages of the secretory cycle are similar to those found with the biochemical method.The lipase granules in the acinar celIs give strong positive reaction in the resting animals.They disappear after the injection of pilocarpine,and are fully restored after 12 hours of recovery.

本文介绍辣根过氧化物酶标记抗体对肝细胞内乙型肝炎表面抗原(HBsAg)的免疫电镜定位方法。电镜样品制备,可采用酶标免疫显色组织切片的再包埋、或直接利用超薄切片进行酶标免疫显色定位。对实验中应注意的一些问题进行了讨论。在电子显微镜下初步观察了HBsAg 阳性肝细胞中HBsAg 的分布和形态,并发现在光学显微镜下,辣根过氧化物酶标免疫定位HBsAg 完全为阴性部位的肝细胞中,有的细胞胞浆内或扩大的内质网池内,尚可见到少量散在的HBsAg 与酶标抗体的沉积物,提示电镜观察有更好的灵敏性。

Radix Trichosanthis injection,in addition to its use as an abortifacient drug of mid-pregnancy,is also used to abort early pregnancies.The inhibitory effect of this drug onthe deciduoma was considered to be an important mechanism of abortive effect in earlypregnancy.It is well known that the regulatory effects of sex hormones on the decidual re-action are related not only to the blood concentrations but also to the amount of hormonalreceptors in the target organs and to the affinity constant(K_A)between the receptors andthe sex hormone.The effects of Radix Trichosanthis injection on the progesterone receptor(PgR)andestradiol receptor(ER)of the deciduoma of pseudo-preganant mice were studied,using aradioreceptor assay which measured the cytoplasmic PgR and ER of the deciduoma in termsof fmol per mg protein.The corresponding affinity constants(K_A)were then obtained bymeans of Scatchard plots.It was found that,after a subcutaneous injection of Radix Trichosanthis 5 mg/kg onthe 7th day of pseudopregnancy,the weight of the deciduoma showed no marked decreasewhen compared with that of the untreated group,and neither did the amount of cytoplasmicER decrease,although the amount of cytoplasmic PgR did decrease significantly.Aftertwo successive subcutanious injections of the aforesaid dose of Radix Trichosanthis on the6th and 7th day of pseudopregnancy,the weight of the deciduoma and the amounts of cyto-plasmic PgR and ER all decreased significantly.The affinity constant(K_A)of cytoplas-mic ER of the deciduoma,however,was not altered by a subcutaneous injection of RadixTrichosanthis on the 7th day,but after two successive injections of drug on the 6th and7th day of pseudopregnancy,this variable declined slightly and maintained the level of10~9M~(-1).By contrast,the K_A of cytoplasmic PgR showed a significant decrease and thisKA was so much reduced as to be hardly measurable,proportionately to the dose of RadixTrichosanthis injection.These experimental results suggest that the inhibitory effects of Radix Trichosanthison cytoplasmic PgR and ER of the deciduoma and the diminution of the affinity constants(K_A)of the two sex hormonal receptors,especially that of cytoplasmic PgR,by Radix Tri-chosanthis bring about the disordered regulation of the decidual reaction by sex hormones.This may be the important mechanism of the inhibitory effect on the deciduoma of RadixTrichosanthis injection.

本文采用放射受体法测定蜕膜瘤胞浆内孕酮受体和雌二醇受体,以fmol/mg 蛋白表示其量,并作Scatchard plot 求其结合常数(K_A)。观察天花粉对假孕小鼠蜕膜瘤孕酮受体和雌二醇受体的影响。在假孕第7天皮下注射天花粉5毫克/公斤/日,蜕膜瘤重量及胞浆内雌二醇受体量及其K_A 与对照组相比无显著差异,但胞浆内孕酮受体量及其真K_A 值则已显著下降。在假孕第6、7二天连续皮下注射同量天花粉,蜕膜瘤重量、胞浆内孕酮受体量、雌二醇受体量及其K_A 均显著下降。而孕酮受体的K_A 值已难以测出。上述结果提示,天花粉减少蜕膜瘤胞浆内孕酮受体和雌二醇受体,并使受体的K_A值(尤其是孕酮受体K_A)变小,从而减弱了激素对蜕膜反应的调节作用,这可能是天花粉抑制蜕膜瘤作用的一重要机制。
