Objective To probe the variation of p53 gene transcription and expression in programmed cell death(PCD) induced by hyperthermia in colorectal carcinoma cell lines and to investigate the clue of hyperthermia for carcinomas.Methods Several common temperatures of hyperthermia(that is,40℃,41.5℃,43℃ and 44.5℃) were chosen in this study,after the PCD was induced by these different temperatures,the comparative analyses on p53 protein expression and mRNA transcription were conducted in HT-29 and Lovo colon carcinoma cell lines having different types of p53 genes by immunohistochemistry and in situ hybridization.Results After 30 min of 43℃ treatment,the p53 mRNA transcription in both cells all increased.While the p53 protein expression in HT-29 cells also increased obviously.Compared with the control group,there was no difference under 41.5℃ hyperthermia for 60 min.No p53 protein was found in Lovo cell lines consistently.Conclusion It is shown that the p53 gene actually participates in the PCD induced by hyperthermia.However,different p53 genes can play different roles in the process.

目的 探讨 p5 3基因在高温诱导大肠癌细胞程序化死亡过程中的转录表达变化 ,为高温治疗肿瘤提供分子生物学线索。方法 选择几种常用高温治疗温度诱导大肠癌细胞呈现PCD特征 ,应用免疫细胞化学和细胞原位杂交技术对含有不同类型 p5 3基因的HT - 2 9、Lovo两种大肠癌细胞p5 3蛋白表达和 p5 3mRNA转录进行了对比分析。 结果 经 43℃处理 30min后 ,两种细胞p5 3mRNA转录均有所增强。HT - 2 9细胞p5 3蛋白表达经 41.5℃处理 6 0min后 ,与对照组比较差异无显著性 ,但经 43℃处理 30min后明显增强 ,Lovo细胞 p5 3始终不能检出。 结论 p5 3基因在高温治疗诱导大肠癌细胞PCD过程中起着重要作用 ,但是 ,不同类型的p5 3基因发挥着不同的作用。

Objective To explore the effects of high insulin and high glucose concentrations on glucose transport activity,the expression of insulin signaling peptides and glucose transporter 4(GLUT4) translocation in primary cultured rat adipocytes.Methods Isolated rat adipocytes were cultured for 24h at insulin(10 4μU/ml) and different concentration glucose(5,10,15 and 25mmol).Then the glucose uptake,cellular contents of insulin receptor substrate(IRS) 1/2,phosphatidylinositol 3-kinase 85 subunit(p85),protein kinase B(PKB) and GLUT4 were measured by Western blotting method.Results These adipocytes treated with insulin and different high concentration glucose had shown to impair glucose uptake in a dose-dependent manner,inhibited cellular IRS1 expression,impaired IRS2 protein expression independent on glucose concentration in the medium,it did not influence the contents of p85,PKB and GLUT4,but decreased GLUT4 translocation.Conclusions Chronic high insulin induced insulin resistance may be caused by the increasing glucose concentration surrounding the cells.The mechanism may be involved in affecting IRSs protein expression and GLUT4 translocation.

目的 探讨高浓度胰岛素和高浓度葡萄糖共同作用对原代培养老鼠脂肪细胞的糖转运活动、细胞内胰岛素信号肽及葡萄糖转运子 4 (GLUT4 )易位的影响。方法 分离的老鼠脂肪细胞在胰岛素 ( 10 4 μU/ml)和不同浓度葡萄糖 ( 5、10、15、2 5mM)培养基中孵育 2 4h ,然后测定糖的转运活动 ;用Westernblot方法测定这些细胞内胰岛素受体底物 (IRS) 1/ 2、肌醇磷脂 - 3 -激酶 85亚单位 (p85)、蛋白激酶B(PKB)和GLUT4的蛋白表达。结果 胰岛素和不同高浓度葡萄糖培养 2 4h ,以一种剂量依赖的方式诱导糖摄取率的减少 ,抑制了IRS1蛋白表达 ,而不影响p85、PKB和GLUT4的表达 ,但抑制了GLUT4易位至质膜 ;胰岛素加正常或不同高浓度葡萄糖治疗 ,均抑制了IRS2蛋白表达。结论 慢性胰岛素治疗诱导的胰岛素抵抗与周围环境中葡萄糖浓度增加有关 ,可能影响了胰岛素受体底物蛋白表达以及GLUT4易位等。

Objective To study the apoptosis of non small cell lung cancer (NSCLC) cells after bronchial artery infusion chemotherapy (BAI) alone or/BAI combined with intrabronchial brachytherapy (IBB). Methods The apoptotic level of lung cancer cell was detected in 55 cases of NSCLC samples by TUNEL. 20 of which were treated with surgical resection alone (group A), 20 of which were treated with BAI alone before surgical resection (group B), and 15 of which were treated with BAI combined with IBB before surgical resection (group C). Results 1. The apoptotic index (AI) in the three groups was 8.92± 2.82%, 10.76±2.18%, 12.43±1.82% respectively with statistical difference (group A vs group B and group B vs group C showed P <0.05, group A vs group C showed P <0.001); 2. AI of squamous carcinoma cells among the three groups showed P <0.05; 3. AI of adenocarcinoma between group A and group C showed P <0.05, and the other comparisons showed P >0.05; 4. The comparisons of AI between squamous carcinoma cell and adenocarcinoma cell in the three groups all showed P >0.05. Conclusions Apoptosis is another important mechanism leading to cell death in NSCLC after BAI alone or/BAI combined with IBB. Apoptosis induced by the latter is higher to BAI alone.

目的 研究支气管动脉化疗及其与支气管腔内放疗联合治疗对非小细胞肺癌细胞凋亡的影响。方法 抽取 1996~ 1998年间在我院接受手术治疗的 5 5例非小细胞肺癌。其中单纯手术切除的2 0例为A组 (对照组 ) ;术前 2周接受过 1~ 2次支气管动脉化疗的 2 0例为B组 ;术前 2~ 3周接受过支气管动脉化疗与支气管腔内放疗联合治疗的 15例为C组。应用原位DNA断端标记法检测 3组标本的癌细胞凋亡水平 ,以光镜下计数的每 10 0个癌细胞的平均凋亡数 (调亡指数 )表示。结果  1.A、B、C 3组间凋亡指数两两比较差异均有显著性 ;2 .3组间鳞癌凋亡指数比较差异有显著性 (P <0 .0 5 ) ;3.3组间腺癌凋亡指数比较 ,A、C两组间有统计学意义 (P <0 .0 5 ) ,其余两组间均无统计学意义 (P >0 .0 5 ) ;4.3组内鳞癌和腺癌间凋亡指数比较均无统计学意义 (P >0 .0 5 )。结论 支气管动脉化疗及其与支气管腔内放疗的联合治疗均能诱导非小细胞肺癌细胞凋亡 ,其中以联合治疗的诱导作用较强。
