We developed a defective herpes simplex virus (HSV) vector system that permitted the introduction of virtually any gene into mammalian central nervous system neurons. The HSV-1 amplicon plasmid, pHSL, contained a βgalactosidase transcription cassette directed by HSV-1 IE68 promoter and two elements from HSV-1 genome: the origin of HSV-1 replication and its packaging site sequence. Being transfected into immortalized cells such as Vero cells in the present of HSV-1 to provide the helper virus function, (in our tests, a temperaturesensitive mutank tsK strain with permissive temperature at 31℃ was used), the prototype vector pHSL could be packaged into HSV-1 particles as a headtotail concatemer with the size of about the genome of HSV-1. A mixed HSV-1 stocks obtained were called dvHSL. Infection of cultured embryonic spinal cord motor neurons and forebrain cortex neurons of rats with dvHSL showed that βgalactosidase was expressed up to twelve days. Via corneal inoculation, the trigeminal ganglia of rats were infected by dvHSL and the expression of βgalactosidase could be detected there at different times after inoculation for at least two months. Infection of the forebrain cortex of rats with dvHSL through stereotactic injection showed that the expression of βgallactosidase was restricted to the injection site for up to two months.

本研究利用构建的Ⅰ型单纯疱疹病毒(HSV-1)扩增子质粒载体pHSL,在辅助病毒HSV-1tsK(许可温度31℃)的辅助下,以首尾相连的连接体形式被包装成复制缺陷型的HSV-1假病毒颗粒,得到一种HSV-1混合毒种dvHSL。将dvHSL接种体外培养的大鼠胚胎脊髓运动神经元和大脑皮质神经元后均可获得报告基因的表达;接种大鼠角膜后,lacZ基因在三叉神经节中持续表达两个月;直接注射接种到大鼠前脑皮质后,可在注射部位的局部表达报告基因近两个月。研究结果表明上述缺陷型单纯疱疹病毒载体可作为神经系统基因转移的工具,应用于神经系统生理、病理及神经系统疾病基因治疗的实验研究

o evaluate the role of keratoepithelioplasty(KEP)for inhibition the corneal superficial neovascularization after alkali burn.Experimental eyes were injured at the circumferential limbal zones by alkali.KEP was performed on the upper limbus with autogeneous lenticules immediately.Examination was taken up to 5 weeks.Results:64% of the transplantative area of the corneas were no or much less new vessels,and the regenerated epithelium was proved histologically to be corneal epithelial phenotype.While,95% of the corneas in controls were neovascularization.The epithelium of them were conjunctival epithelium,some goblet cells were seen in it.Conclusion:This study suggested that KEP played a important role in blocking the ocular surface neovascularization and reconstructing the corneal epithelial integrity after alkali burn....

为评价角膜上皮移植术对角膜碱烧伤后表层新生血管的抑制作用,对大鼠角膜全周碱烧伤后立即行上缘自体角膜上皮移植术,连续观察5周。结果,64%移植区角膜表面无或有很少新生血管,组织学证实为角膜上皮表型。而95%的对照组角膜表面全部新生血管化,组织学显示有散在杯细胞为结膜上皮。本文提示角膜上皮移植术对碱烧伤后重建角膜表面的完整性,并预防角膜新生血管化,具有重要价值。

In order to observe the suppressive effect of anti-CD 25 monoclonal antibodies (ART-18) on corneal graft rejection, the model of SD rat to Lewis rat penetrating Keratoplasty was applied, and were treated with anti-CD 25 McAb after operation. The results showed that the occurence rat of corneal rejection was 100% in the control group, it was 53.3% in the group of rat treated with ART-18 and was 26.7% in the group of rat treated with both ART-18 and Cyclosporin A. These results indicated that anti-CD 25 monoclonal antibody suppressed the corneal graft rejection effectively, and ART-18 combined with Cyclosporin A showed much better effect.

为了观察抗CD25单克隆抗体(ART-18)对角膜移植排斥反应的抑制作用,应用SD大鼠对Lewis大鼠进行穿透性角膜移植,术后用抗CD25McAb治疗。结果显示:对照组大鼠角膜排斥反应发生率为100%;ART-18治疗组角膜排斥反应发生率为53.3%;ART-18联合环孢霉素A治疗组角膜排斥反应发生率为26.7%。结果表明:抗CD25McAb能够有效抑制角膜排斥反应,其中抗CD25McAb联合环孢霉素A治疗效果更好。
