The studies on suitable conditions for pullulan fermentation according to the results obtained from the shaking flask tests was carried out in 16L auto-controlling fer-mentor. It fount that the optimal DE value of starch hydrolyzate was 40-50 when 10% starch was used as carbon source. The optimal concentration of ammonium sulfate in the medium for fermentor was different from that of the shaking flask. The fermentation kinetics and effects of seed age, seed volume, airflow rate, pressure of tank, agitation speed...

在摇瓶发酵条件研究的基础上。于16L自控发酵罐上进行了罐上发酵条件优化研究,发现以10%淀粉水解物为碳源时,淀粉水解物的最适DE值为40—50,发酵培养基中的硫酸铵最适用量不同于摇瓶发酵时的量,种龄和接种量、通气量、罐压、搅拌速度和搅拌叶轮挡数等均对多糖的产生有较大的影响。另外还进行了发酵过程的动力学的研究。

Xanthomonas campestris ZZ 1 was used as production strain to study the optimization of fermentation conditions of xanthan gum in shaking flaves.And the optimum fermentation conditions were as follows:pH 7.2,CaCO 3 0.3%, temperature 30℃,contents of medium in flasks 20%.viscosity of finished products were 1 400 cp,which increased by 16.53% compared with the basic culture conditions.

以甘蓝黑腐病黄单胞菌ZZ 1(XanthomonascampestrisZZ 1)为生产菌株 ,进行了摇瓶条件下黄原胶发酵条件的优化研究 ,得到最佳摇瓶发酵条件为pH 7.2 ,CaCO3 0 .30 % ,培养温度 30℃ ,装料量为 2 0 % ,最终成品粘度可达 14 0 0cp ,较基本培养条件提高了 16 5 3% .

The corynebacterium glutamicum strain JSIM201 was selected as the starting strain. By treatment of UV ray and EMS, an uracil auxotrophy mutant strain U12 was obtained. Using glucose and ammonium sulfate as the carban source and nitrogen source, the mutant could accumulate an ultraviolet absorption substance in its cultural broth. 以谷氨酸棒杆菌JSIM 201为出发菌株,通过紫外线和甲基磺酸乙酯诱变处理,得到了一株尿嘧啶营养缺陷型突变体U 12菌株,它能以葡萄糖为碳源,硫酸铵为氮源,在发酵液中积累一种紫外吸收物质.该紫外吸收物质经物理、化学分析鉴定,证明是乳清酸.对U 12菌株进行了发酵条件优化研究,在最佳发酵工艺条件下,积累乳清酸最高达到8.6g/L.
