A new method for constructing nonra-dioactive hybridization probes was studied,which allowed crosslinks to be producedbetween HRP and single-stranded DNA.Such single-stranded DNA-HRP complexeshave been tested for blot hybridization.After hybridization and incubation witha substrate solution, sequences comple-mentary to the probe could be visualizeddirectly without the need of tedious cyto-chemical sandwich methods. This proceduremight detect target DNA in the 1-4 pgrange.

本文叙述一种将辣根过氧化物酶(HRP)与单链DNA交联以制备非同位素核酸杂交探针的新方法。HRP-单链DNA复合物可用于斑点杂交,经杂文并与HRP底物溶液作用后,可直接检出互补的DNA靶序列,而不需繁冗的细胞化学夹心法。本法可检出 1~4 pg的靶DNA。
