Electronic structure of 23 platinum (Ⅱ) complexes of the types of [(NH_3)_2Ptx_2], [(CH_3NH_2)_2Ptx_2], [enPtx_2] and [1,2-(NH_2)_2C_6H_(10)Ptx_2] (X=Cl-,Br~-, I~-,1/2(oxalate)~(2-),1/2(malonate)~(2-), 1/2(hydroxymalonate)~(2-), 1/2(methylmalonate)~(2-),1/2(ethylmalonate)~(2-),1/2(1,1-cyclobutanedicarboxylate)~(2-), SCN~-)have been calculated by CNDO/2-SCF method. Seven regressions are obtained from the calculated electr onic indices according to biological activity. The results show that the toxicity and activity of the complexes are closely related to both the amine and the acid redical ligands. It seems that the toxicity of a Pt complex is decreased when appropriate cyclo-amine and organic acid radical serve as ligands, and a high electron density of N atom increases the activity of the complex.

本文用CNDO/2方法计算了[(NH_3)_2PtX_2],[(CH_3NH_2)_2PtX_2],[enPtX_2]和[1,2—(NH_2)_2C_6H_(10)PtX_2]四个系列的23种配合物的电子结构,并用配合物的抗癌实验指标与由量子化学计算所得结果进行线性回归,得到七个回归方程,讨论了配合物的结构和活性间的关系。结果表明,合成一种毒性低、剂量小、活性高的配合物,不但要考虑胺配体对配合物抗癌活性的影响,而且酸根配体对配合物的活性也起着重要的作用。

In a previous article we have reported xylene cyanol (XC), one of the two dyes selected from 70 species of acid dyes for the determination of amines. These dyes possessing sulfonic acid or carboxylic acid radical have not yet been reported in the literature. This article reports erioglaucine(EG), the other best dye selected. The limits of detection of this dye for benethtropine [-1,1-diphenyl-2(3-tropanyl) ethane] and benactyzine (benzilic acid 2-diethylamino-ethyl ester) are 15 ng/ml and 20 ng/ml respectively....

In a previous article we have reported xylene cyanol (XC), one of the two dyes selected from 70 species of acid dyes for the determination of amines. These dyes possessing sulfonic acid or carboxylic acid radical have not yet been reported in the literature. This article reports erioglaucine(EG), the other best dye selected. The limits of detection of this dye for benethtropine [-1,1-diphenyl-2(3-tropanyl) ethane] and benactyzine (benzilic acid 2-diethylamino-ethyl ester) are 15 ng/ml and 20 ng/ml respectively. The absorption coefficients (E_(1cm)~(1%)) of the EG-amine complexes are 4~6 times as high as those of bromothymol blue (BTB). Chloroform extract of the complex shows greenish blue color, λ_(max), 635.5 nm. For the determination of amines the preferable concentration of the dye used is 2 × 10~(-3) mol/L. The calibration curves are linear in the range of 0~7(I)and 0~10(Ⅱ)μg/ml. With the dye concentration of 2×10~(-3) mol/L, the mole ratio of dye-amine complex in the chloroform extract for benactyzine is EG : benactyzine=1:1, and 0.5:1 with 2×10~(-4) mol/L. But in the ease of benethtropine, the mole ratio with different dye concentration keeps constant at 1.3:1(dye:amine). The coefficients of variation for determination of 0.2 μg/ml benethtropine and benactyzine are within±5%, and±1~2% for 2.5μg/ml,5.0 μg/ml and 10.0 μg/ml. Similar to XC, this dye belongs to a new type of acid dye for the determination of amines, and the following portion is also considered to be the fundamental structure:■

前报报道了自70种色素中研究选出测定胺类的两种色素之一:二甲苯蓝(XC),本文报道另一种色素:羊毛罂粟蓝(EG)。经研究得出:EG对亲脂性较大的胺,苯乙托品(Ⅰ)的检出限为15ng/ml,苯乃辛(Ⅱ)为20 ng/ml,其吸收系数(E_(1cm)~(1%))(Ⅰ为36.89×10~2,Ⅱ为24.36×10~2)为溴百里酚蓝(BTIJ)的4.6~5.9倍。EG—胺配合物的氯仿液呈绿蓝色,λ_(max)635.5nm。测定时色素浓度应为2×10~(-3)mol/L。检量线在0~7(Ⅰ)与0~10(Ⅱ)μg/ml为直线。EG—胺配合物的分子比在EG浓度为2×10~(-3)mol/L时,EG:Ⅱ=1:1;2×10~(-4)mol/L时则为0.5:1。Ⅰ则在EG浓度不同时分子比基本不变,为1.3:1。用EG测定Ⅰ或Ⅱ0.2μg/ml时,CV在±5%以内;2.5,5.0,10.0μg/ml时为±1~2%。EG与XC一样,为文献上尚未用来测定胺类的新类型酸性色素,属同一类,其基本构造为:■

Accordingly, the authors found another dye of this new type, i. e. patent blue V(PBV). Chloroform extract of the complex of PBV-amine shows greenish blue color, λ_(max) 632 nm. The preferable concentration of PBV used for determination of amine is 4×10~(-3)mol/L. The limits of detection of this dye for benethtropine(Ⅰ) and benactyzine(Ⅱ) are 16 ng/ml and 37 ng/ml respectively. The absorption coefficients (E_(1cm)~(1%) of these dye-amine complexes [37.6×10~2(Ⅰ) and 22.0×10~2(Ⅱ)] are4.1~6 times as high as those of bromothymol blue (BTB), but for more hydrophilic amines, E_(1cm)~(1%) are lower than those obtained from BTB. The calibration curves are straight lines in the range of 0~6.5 μg/ml for Ⅰ and 0~7.5μg/ml for Ⅱ. With PBV concentration of 4×10~(-3)mol/L, the mole ratio of dye-amine complex in the chloroform extract for Ⅱ is PBV: Ⅱ=2:1 and 1:1 with 4×10~(-4)mol/L. In the case of Ⅰ, the mole ratio with different dye concentration (4×10~(-4)~4×10~(-3)mol/L) keeps constant at 3:1 (dye: amine). The coefficients of variation for determination of 0.2 μg/ml of Ⅰ and Ⅱ are within ±5%, while ±1~2% for 2.5 μg/ml, 5.0μg/ml and 10.0 μg/ml. PBV has the same properties and sensitivity as xylene cyanol (XC) and erioglaucine (EG). On the basis of the studies of three dyes, namely XC~(1), EG~(2) and PBV, it is further considered that the following moiety is the fundamental structure of this new type acid dye:

前报认为此新类型蓝色酸性色素的基本构造为二氨基三苯甲烷磺酸,据此,找出了另一种同类型的酸性色素——专利蓝Ⅴ(PBV)。对它进行了配合物的吸收峰、试剂空白的降低、试剂浓度的选定、最佳pH值的选择、检出限、检量线、色素与两种胺的分子配合比、PBV测定法的精密度、五种胺配合物的吸收系数比较等研究,证明它与二甲苯蓝、羊毛罂粟蓝有类似的性质以及同样的灵敏度。进一步说明所认定的此类型的基本构造是对的。
