The construction in vitro of hybrid plasmids of EcoRI-linearized pla- smid pUB110 of B.subtilis and E.coli plasmid pBR322 digested by EcoRI, was described and named pHE1-a and pHE1-b reshectiuely. Under eletron microscopy, they were found to be covalently closed circular DNA molecules. The maps obtained by analyzing with restriction enzymes EcoRI, BamH I, Bgl Ⅱ, Xba I, Hind Ⅲ and Pst I indicated that these two recombinant plasmids were similar in size but in opposite linking orientation. The detection of resistance to antibiotics of strains transformed revealed that the ampicillin and tetracycline resistance from E.coli could not be expressed in B.subtilis, but kanamycin and neomycin resistance carried by B.subtilis could be expressed in both hosts. Since these two hybrids are replicatable in both E.coli and B.subtilis, they can be used as cloning vectors in both bacteria.

Isolation of antibiotic resistant mutations is a useful approach to study structure and function of ribosome. Both spiramycin and ribostamycin inhibit protein synthesis on ribosome. In this paper nineteen SpiR mutants and fourty-nine. RibR mutants were isolated. Five out of these mutants showed a detectable ribosomal protein with altered migration on the two-dimensional polyacrylamide gel. The altered proteins were S4, S5, L18, L23 and L13, respectively. They, however, were not responsible for SpiR or RibR resistant phenotype. Both spi and rib were mapped in the main ribosomal protein gene cluster near the origin of chromosome. The order of these loci was cysA-rib-rpsL-spi-ts-5-rpsC-rpsE-ts. The rplW was in rpsL-rpsE cluster, but rplR was not in this region. The macromolecular synthesis of mutant SP4 which carried altered protein L18 was lower than that of the control strain. The assembly time and functional capacity of the mRNA wetre determined by pulse-labeled experiments. It was foiund that the RNA synthesis of strain TDL23 which had only an altered protein L23 was reduced, but the protein synthesis of the strain was similar to the control strain. It was shown that the capacity of the mRNA to synthesize protein in strain TDL23 was increased as compared to the control.

从湖北省两种土壤中分离获得11株快生型大豆根瘤菌。在YEM培养基上产酸,世代时间低于4小时。这些菌株除对柠檬酸盐等五种碳源不能利用外,对供试的甘露糖等16种碳源均可利用。除一个菌株外,其余的10个菌株均能耐1%NaCl,但不超过2%。用链霉素等9种抗菌素定量加入YEM培养基;培养分离出的菌株,根据其抗性反应,可将它们分为5个抗菌素抗性类型。
