A simple and specific method for estimation of free proline in human plasma was recommended in this paper. The proline was separated completely from other amino acids with n-butanol-aoetie aoid-ethanol-water(5:1:2:1) as mobile phase by paper chromatography. The chromatograms were run overnight for a total of 30 hr. at room temperature. They were sprayed with isatin and heated at 80℃ for 30 min, and then a dark blue spot was manifested. A simple and specific method for estimation of free proline in human plasma was recommended in this paper. The proline was separated completely from other amino acids with n-butanol-aoetie aoid-ethanol-water(5:1:2:1) as mobile phase by paper chromatography. The chromatograms were run overnight for a total of 30 hr. at room temperature. They were sprayed with isatin and heated at 80℃ for 30 min, and then a dark blue spot was manifested. The excess of isatin was washed with water at 40℃ for 5 min, and then the chromatographio paper was dried at 60℃. The blue spot was cut off and put into 5 ml phenol saturated with water. Eluted for 13 min, it was determinated at 608℃1nm by UV-speotrophotometer.It was found that the mean plasma concentration of free proline in normal subjects as 235,4×74.4 μmol/l (n = 36) and 107.8×39.3μmol/l in late stage of schistosomiasis (n = 22). An apparent difference was observed between normal subjects and patients (p<0.01).This method has advantages of good specificity, high recovery (mean value 97.47%) and simple experimental apparatus; therefore, the method is particularly adapted to clinical diagnoses of hepatic cirrhosis.

本文报道一种测定人血浆游离脯氨酸的简便方法。用纸层析法分离,以正丁醇:冰醋酸:乙醇:水(5:1:2:1)为展开剂,吲哚醌试液为显色剂,生成蓝色化合物于分光光度计608±1nm处测定其吸光度,通过标准曲线计算其含量。用本法测得正常人血浆游离脯氨酸量为235.4±74.4μmol/L(n=36),晚期血吸虫病人为107.8±39.3μmol/L(n=22),两者有显著差异(p<0.01)。本法专一性强,回收率高(97.47%),检出限为0.5μg,所用仪器设备简单,适合于临床检验用。

由嗜热脂肪芽胞杆菌(Bacillus stearothermophilus SIPI1.687)的粗提取液,经液-液双相抽提、DEAE-Toyopearl 650 M 离子交换柱层析及 SIPI-Ⅱ蓝色(?)料配基层析等分离提纯,可得甘油激酶(81U/mg),PAGE 鉴定为单带。用 Sephacryl 400凝胶过滤测得分子量为209,000道尔顿,SDS-PAGE 测定结果表明该酶由四个分子量均为50,000道尔顿的相似亚基组成。酶作用最佳 pH 9.4,pH 5.8~11.2时稳定;最佳温度为50℃,有较好的热稳定性。以甘油为底物时米氏常数为6.25×10~(-5)M。
