本课题为1989~1991年国家自然科学基金、浙江省自然科学基金资助项目及浙江省教委研究项目。本文是对舟山及南麂海域蔓足类研究的总结。生态学研究包括:种类组成与区系性质,生态分布,种群生态和群落生态,形态、结构与环境以及行为生态;生物学研究包括:繁殖、附着与生长,雄性生殖腺的组织学、组织化学和亚显微结构以及藤壶的食性。

Objective:To develop a sensitive,specific and accurate method(reverse transcription polymerase chain reaction enzyme linked sandwich hybridization) for quantitative detection of human FGF mRNA.Methods:Two specific probes were designed and their sequences could respectively match different regions of the product of mRNA. Objective:To develop a sensitive,specific and accurate method(reverse transcription polymerase chain reaction enzyme linked sandwich hybridization) for quantitative detection of human FGF mRNA.Methods:Two specific probes were designed and their sequences could respectively match different regions of the product of mRNA. One is the capture probe which has amino modified at its end and can couple with the N oxysuccinimide esters(NOS) group on the DNA binding well through covalent attachment. The other is detector probe which is biotinylated. Extracted total RNA from human peripheral blood mononuclear(PBMC) and it was amplified by RT PCR. Heat denatured PCR products and detection probe were added to the wells having coated with capture probe and then sandwich hybridization occurred between capture probe,target amplicon and detection probe. After the addition of avidin horseradish peroxidase and its substrates,optical density was read at 450 nm.Results:The developed technology for the detection of human FGF mRNA is good in sensitivity and specificity. No positive signal was observed in enzyme linked sandwich hybridization with non target amplified products and the reproducibility was good.Conclusion:With its simplicity,sensitivity and specificity,the technology of RT PCR and enzyme linked hybridization microplate could be an alternative method for the assay of FGF mRNA.
