At 75.5℃ the kinetic constants of thermal in activation for immunochemical activity are as follows: the rate constant of inactivation (k) 7.29×10~(-5) sec~(-1), the free energy of activation (E~+) 95.4 KCal·Mole~(-1), the enthalpy of activation (△H~+) 94.7 K Cal·Mole~(-1), and the entropy of activation (△S~≠) 193 Cal·Mole~(-1). Degree~(-1). From evidence furnished by SDS-polyacrylamide electrophoresis it appears most likely that both foetal AFP and cancerous AFP consist of single polypeptide chains. Their amino acid compositions are very much alike and their S-S bonds play a very important role in maintaining the immunochemical activity of AFP. Both GNBr-cleft AFP samples give 7 main peptide-fragments and corresponding peptides have closely similar molecular weights. From the above mentioned results it may be concluded that no differences in the nature of these two AFP's have been observed.

Furthermore, the physicochemical and biological properties of the two main components, 1588-74-3 and 1588-74-9 (amounting to 23.4% and 11.3%, respectively), were studied.The paper and thin-layer chromatograms of the acid hydrolysate of these two main components were different from those of bleomycin A_2 and A_5. In addition, these two main components of antibiotic 1588 were also found to be different from other members of the phleomycin-bleomycin group in their UV absorption ratios, pH stabilities and the behavior of chromatography. So, antibiotic 1588 might be considered as a new member of this group.

The production of the chymotrypsin inhibitor by Streptomyces S-81-24 reached a maximum after two to three days in submerged culture. Chymotrypsin inhibitor can be extracted either by absorption using D-resin or with n-butanol at pH 2~3. It is soluble in water and methanol, but insoluble in non-polar solvents, such as, ethyl-acetate, ether, benzene, chloroform etc. Novisible absorbtion is observed in U. V. spectrum. It inhibits not only chymotrypsin strongly but also inhibits trypsin and pepsin to a certain extent....

链霉菌S-81-24所产生的糜蛋白酶抑制剂,其高峰期是在摇瓶培养的2~3天。提取时既可以利用大孔树脂吸附也可以用正丁醇在pH2~3进行萃取。它易溶于水、甲醇,但不溶于乙酸乙醋、乙醚、苯和氯仿等非极性有机溶剂。没有明显的紫外吸收峰。初步测定,它不仅对糜蛋白酶有强的抑制作用而且对胰蛋白酶、胃蛋白酶也有一定的抑制作用。但是这种多功能性是来自样品的同一组分还是不同组分尚待今后样品进一步纯化后再行深入探讨。
