The cytoplasmic postive expression of CK, AFP, S-100 protein and Vimentin of epithelial tumor cells were found in 14,10, 9 and 6 cases respectively. positive staining of CEA, P53 and P16 of nuclei of epithelial tumor cells were found in 11, 9 and 7 cases respectively. Among the 12 cases who underwent surgical exci-sion, 8 patients with fetal type survived over 5 years. Conclusions This study suggests that immunoreac-tion of hepatoblastomas are different in the histological subtypes. This expression may correate with the dif-ferentiation ans prognosis of the tumors.The prognosis of fetal type is better than the other histologic sub-types.

目的探讨肝母细胞瘤各组织类型的免疫表达和预后的关系。方法对24例肝母细胞瘤进行临床病理分析和组织学分型,其中18例作7种标记的免疫组化研究。结果胎儿型12例,胚胎型7例,间变型3例和上皮间叶混合型2例;细胞角蛋白、甲胎蛋白、S-100蛋白和波形蛋白在肿瘤上皮细胞浆的表达分别为14例、10例、9例和6例,癌胚抗原、P53和P16蛋白在细胞核中的表达分别是11例、9例和7例;手术切除肿瘤12例中8例生存期超过5年者均为胎儿型。结论肝母细胞瘤不同组织类型的免疫表达各异,并与肿瘤的分化和预后有关,胎儿型预后优于其他类型。

目的 探讨小儿肝母细胞瘤的临床病理特点及预后。方法 对 2 4例小儿肝细胞瘤进行临床病理分析和组织学分型 ,其中 18例作 7种标记的免疫组化研究 ;对手术病例进行随访观察。结果 上皮型肝母细胞瘤 2 2例 ,其中胎儿型 13例 ,胚胎型 7例和间变型 3例 ;上皮间叶混合型 2例。细胞角蛋白、甲胎蛋白、S- 10 0蛋白和波型蛋白在肿瘤细胞浆的表达分别为 14例、10例、9例和 4例 ;癌胚抗原、p5 3和 p16蛋白在肿瘤细胞核的表达分别为 11例、9例和 7例。手术完整切除肿瘤 12例中存活 10例 ,8例生存期超过 5年。所有存活病例均为胎儿型。结论 肝母细胞瘤可分为若干组织类型 ,不同类型的免疫表达各异 ,组织类型和预后有关。对胎儿型肝母细胞瘤 ,只要早期诊断和完整切除 ,是可能完全治愈的。

Objective To analyze epithelial-mesenchymal transition (EMT) in HPV16 E6 and E7 immortalized oral epithelial cells and biological characteristics of EMT cells. Methods We established a human immortalized oral epithelial cell line (HIOEC) by transfecting normal oral epithelial cells with recombined retroviral system LXSN HPV16 E6 and E7. The phase-control microscope and differential interference microscope were used to observe the morphology of HIOEC cells. Half-quantify the transcription of snail E-cadherin, β-catenin, wnt-4 and wnt-5a in normal cells and HIOEC cells with RT-PCR. Identify the expression of cytokeratin and vimentin in normal cells and HIOEC cells with histoimmunochemical staining. Observe the distribution of Actin filament cytoskeleton of normal cells and HIOEC cells with immunofluorescence staining under CONFACAL. Results HIOEC cells were immortal but not tumorigenic. Cells retained many characteristics of keratinocytes but displayed spindle-like shape and growth spreaded.The transcription of E-cadherin and wnt-4 were down-regulated while β-catenin and wnt-5a were up-regulated in HIOEC cells. Cytokeratin and vimentin were both positive in HIOEC cells. Actin filaments were more abundant and arranged in bundles in HIOEC cells. Cytoskeleton was much stronger in HIOEC cells than that in normal cells. Conclusion HIOEC cells are under EMT. The cells show partial characteristics of mesenchymal cell.

目的研究人乳头状瘤病毒HPV16E6/E7永生化口腔上皮细胞的上皮—间叶变(EMT),以及发生EMT细胞的生物学特征。方法相差显微镜、微分干涉显微镜观察HPV16E6/E7永生化口腔上皮细胞系HIOEC细胞形态学改变;透射电镜观察细胞间桥粒形成情况;RTPCR比较正常口腔上皮细胞和HIOEC细胞中βcatenin、上皮钙黏蛋白(Ecadherin)、wnt4、wnt5a、snail基因的转录情况;免疫组化检测HIOEC细胞中细胞角蛋白(CK)、波形蛋白(Vim)的表达。免疫荧光、激光共聚焦显微镜观察Actin细胞骨架的改变。结果HIOEC细胞呈梭形成纤维细胞样改变,细胞分散生长;RTPCR结果显示HIOEC细胞Ecadherin、wnt4转录水平下调,wnt5a转录水平上调,βcatenin、snail基因无明显改变。HIOEC细胞共表达CK和Vim。Actin细胞骨架在HIOEC细胞中更显丰富,呈束排列。结论HPV16E6/E7永生化口腔上皮HIOEC细胞发生上皮—间叶变,细胞表现间叶细胞的部分生物学特征。
