During metamorphosis the activity of the hemolymph phosphomonoesterase of Eri-silkworm changes in a regular pattcrn according to the developmental stages. In thebeginning of the last larval instar, on the fourth day of the fifth instar and in the pre-pupal stage the enzymic activity is the highest; it decreases sharply just after pupationand increases again before and after emergence. The enzymic activity calculated perunit volume of hemolymph shows no significant difference between the sexes. However,from the middle of the fifth instar, the enzymic activity calculated per unit weight ofprotein of the males is always higher than that of the females. The distribution andchange of activities of this enzyme between different tissues are as follows: In themiddle of the fifth instar larvae, the enzyme activity in the digestive tract is higher thanthose of other parts of the body, that of the mid-gut being the highest. The lowestvalues of enzymic activity was found in the fat bodies and silk glands. The enzymeactivity in the fat bodies increased in the pupal stage, reached the highest level afteremergence. These facts indicate that in the middle of fifth larval instar the mid-gut, where active phosphorylation and dephosphorylation are carried out, is theactive site of metabolism and that in the pupa and adult stages when the digestivetract degenerates, the fat bodies play an important role in metabolism. The hemolymphof insects differs from the blood of other animals by a high content of phosphates. Thetypes and the amounts of phosphates in the hemolymph are found to be influenced bythe selective bydrolysis in the hemolymph and the selective secretion from the cells, inagreement with the view expressed by Wyatt (1963). During metamorphosis the selec-tive hydrolysis is effected by the presence and changes of activities of the phosphataseswith different specificities. The present study further indicates that the mid-gut and thefat bodies of the fifth instar larvae and the fat bodies of the pupae and adult are themain sources of hemolymph phosphates. The properties of hemolymph acid phospho-monoesterase and the inhibition and activation of enzymic activities by some metal ions,organic acids, etc., have also been studied.

Cellulase "EA_3-867" is an enzyme complex prepared from a Trichoderma viride EA_3-867 mutant, isolated in this institute in 1972.The present cellulase preparation is light brown in appearance. It consists of C_1 and G_x factors, β-glucosidases, pectinase, hemicellulase and traces of other minor components, and exhibits remarkable activity in decomposing cellulose and pectins. A significant amount of intact protoplasts can be obtained from different tissues of various species of plants after incubating them with the cellulase "EA_3-867" solution. Protoplasts obtained from tobacco leaf and callus through incubation with such enzyme preparations have successfully given rise to whole plants at optimum cultural conditions. Our experimental results show that the cellulase "EA_3-867" is an ideal agent for the preparation of protoplasts for plant somatic hybridization and other genetic studies and is apparently comparable to the cellulase "Onozuka" R-10 generally used in many laboratories.

Three species of nitrogen-fixing blue-green algae, Anabaena azotica (HB 686), Anabaenacylindrica and Anabaena 7120, were studied with reference to nitrogenase activity in intactcells as well as in cell-free extracts. Although the nitrogenase activity of intact cells of HB 686(51.9mmC_2H_4/O. D./30min.) was higher than A. 7120 (16.4mmC_2H_4/O. D./30min.), itsactivity in the crude extract was the lowest. This may be due to the abundance of mucilagesin the algal mass. The effects of Mn~(++) and Fe~(++) on the nitrogenase activity of these algae weretudied also, and the reaction kinetics with different enzyme concentrations were estimated. Theresults obtained point out that in A. cylindrica there is a lack of activating cofactor, which hasbeen demonstrated in Rhodospirillum rubrum. Besides, the filaments of Anabaena 7120 weretreated with toluol-ethanol and their N_2-fixing activity in situ was estimated in an attempt tofind out the mechanism of protection from oxygen.
