Partial protection by the competitive inhibitor, dihydrocinnamate suggests that the group affected is at the active site of the enzyme. The reaction of NPAB with proteins was pH dependent. The effect of pH on the inhibition of insulin by NPAB shows that this reaction is controlled by a dissociable group with a pK of 6.5, probably an imidazole group of a histidine residue. Similar results were obtained for ribonuclease, the dissociable group concerned, however, has a pK of 7.6. The effect of pH on the reactions of papain and α-chymotrypsin with NPAB was more complex, it appears probable that more than one dissociable group takes part in the control of these reactions.
