CNBr cleavage followed by peptic digestion released an active fragment from the mung bean trypsin inhibitor. This fragment could be separated by using affinity chromatography with immobilized trypsin and gel filtration on sephadex G-50 and was found to inhibit trypsin stoichiometrically. It appeared to be derived from the domain including the N-terminus of the original inhibitor, whereas the other active site lost its activity during the CNBr cleavage.The active fragment consisted of two peptide chains linked...

This fragment could be separated by using affinity chromatography with immobilized trypsin and gel filtration on sephadex G-50 and was found to inhibit trypsin stoichiometrically. It appeared to be derived from the domain including the N-terminus of the original inhibitor, whereas the other active site lost its activity during the CNBr cleavage.The active fragment consisted of two peptide chains linked by two disulphide bridges, comprising 26 and 9 amino acid residues with Ser and Phe as the Nterminal, Leu and Hse as the C-terminal residues respectively.The complex of the active fragment with bovine trypsin in equal molar ratio could be crystallized in thin plates. At low pH this complex could be dissociated and separated into its original components through gel filtration on Sephadex G-50. In spite of being combined with trypsin for one month or more, the active fragment still retained full activity.The stability of the active fragment was studied at different temperatures over a range of pH3—12. No measurable changes in the inhibitory activity were observed even at 100℃ for 10 minutes when the pH was lower than 6. At higher pHs the inhibitory activity deoreased in a way similar to that of the intact inhibitor.After further digestion with aminopeptidase and carboxypsptidases in excess, the size of the active fragment could be reduced further to about 27 amino acid residues without any loss in actrivity. So far as we are aware this was the smallest active fragment from any known trypsin inhibitor.
