Further, IAA-CO - NH-BSA and IAA-N-OVA were synthesized with1-ethyl-3-(3-dimethylaminopropyl )-carbodiimide or via forrnadehyde respectively.Therefore, two kinds of indirect enzymelinked immunosorbent assays (ELISAI andELISA2) were developed, utilizing IAA-CO-NH-BSA or IAA-N-OVA for coatingwells. The sensitivities of ELISA1 and ELISA2 were 0. 35 pmol and 1. 80 pmol, and the detection ranges were 0. 78-800pmol and 1. 95-2000pmol respectively. The antiserum with high affinity to carbofuran was obtained from New Zealand white rabbits by using synthetic antigen as immunogen. The polyclonal antibodies were separated from antiserum by salting out method and purified by reversed phase adsorption of DEAE cellulose. The horseradish peroxidase(HRP) tagged antibody was prepared by means of modified NaIO 4 method and the HRP tagged hapten was prepared by means of mixed anhydride method. Based on these, the indirect competitive ELISA, antibody or Twenty two points, plus triple word score, plus fifty points for using antigen coated direct competitive ELISAs, which are highly specific to carbofuron, were established. With optimized conditions of ELISA, the linear concentrations ranged from 10 0~10 -4 μg/mL and the detection limit was lower than 0.01ng/mL for the determination of carbofuran. Some common used carbamate insecticides and BFOH didn't interfere with the analysis of carbofuron.

Then two haptens named 4 [〔(2,3 dihydro 2,2 dimethyl 7 benfuranyloxy) carbonyl〕 amino] hexanoic acid(BFNH) and 6 [〔2,3 dihydro 2,2 dimethyl 7 benfuranyloxy) carbonyl〕 amino] butanoic acid(BFNB) were synthesized with the reaction between DDBC and amino hexanoic acid or amino butanoic acid under different conditions. The BFNH and BFNB were conjugated to bovine serum albumin (BSA) with the modified active ester method to form two antigens for producing the high titre antibodies to carbofuran. The ratios of BFNH BSA and BFNB BSA were 10∶1 and 13∶1, respectively. Other two coating antigens for carbofuran, BFNH OVA and BNFB OVA were also synthesized by conjugating BFNH or BFNB to ovalbumin (OVA) with the mixed anhydride method, and their ratios were 4∶1 and 3∶1, respectively.
